Bazı buğday tescilli çeşit ve ileri hatlarının biyokimyasal markörler ile tanımlanması

Makarnalık buğday genotiplerinin farklılıklarının tanımlanması amacıyla yapılan bu araştırmada glütenin proteini moleküler markör olarak kullanılmıştır. CIMMYT'ten sağlanan toplam 18 makarnalık buğday genotipinin sodyum dodesil sülfat poliakrilamid.jel elektroforezi (SDS-PAGE) tekniği ile glütenin bantları elde edilmiştir. Çalışmamızda elde edilen sonuçlara göre, materyali oluşturan genotiplerin tamamı kendilerine özgü glütenin bant desenleri oluşturmuşlardır. Çalışmada kullanılan makarnalık buğday genotiplerinin (buğday çeşit ve ileri hatları) birbirlerinden oldukça farklı glütenin bant desenleri oluşturduğu görülmektedir. Literatür bildirişlerine göre glütenin bant desenleri ile buğday ıslah programlarında yer alan hatların ve çeşitlerin tanımlamaları yapılabilmektedir. Ancak aynı pedigri grubunda yer alan genotiplere ait glütenin bant desenleri incelendiğinde bunların genotiplerde mevcut olan benzerliği ortaya koyamadığı gözlenmiştir.

Identification of some wheat commercial varieties and advanced lines by biochemical markers

The objective of this study was to identify durum wheat genotypes by using glütenin protein as a molecular marker. Glütenin banding patterns of 18 durum wheat genotypes from CIMMTT collections were obtained by using the SDS-PAGE technique. Evaluation of these glütenin banding patterns showed that these genotypes were totaly different from each other and variation among genotypes could be expressed by these patterns. According to the results, it is clear that wheat lines and varieties of breeding programs could be identified with glütenin banding patterns. However, no similarity was observed in the banding patterns of the genotypes in the same pedigree groups.
