Akmut ve ayvada nişasta jel elektroforez tekniğine göre peroksidaz izoenzim analizleri için en uygun yöntemin belirlenmesi
Bu çalışma, armut çeşitleri ile ayva anaçlarında çeşitli amaçlarla kullanılabilecek peroksidaz (PRX) izoenzim analizleri için en uygun nişasta jel elektroforez (SGE) yönteminin belirlenmesi üzerine kurulmuştur. Denemede, Bartlett (BT= Williams) ve Beurre Hardy (BH) armut çeşitleri ile QA ayva anacının yıllık sürgünlerinden alınan kabuk dokusu örneklen kullanılmıştır. SGE için Krebs (1996), modifıye edilmiş Krebs (1996) ve modifıye edilmiş Santamour ve ark (1986) olmak üzere 3 farklı ekstraksiyon solüsyonu; Histidine-citrate, pH 5.7, Morpholine-citrate pH 6.25 ve Lithium. Borate/Tris-citrate pH 8.3 olmak üzere 3 farklı jel ve elektrot sistemi ve Wendel ve Weeden (1989), modifıye edilen Santamour ve ark. (1986) ve Santamour ve ark (1986) Guaiacol yöntemi olmak üzere 3 farklı boyama sistemi denenmiştir. Sonuç olarak, armut ve ayvada SGE ile peroksidaz İzoenzim analizlerinde Santamour ve ark. (1986) yöntemine göre ekstrakte edilen örneklerin Lithium borate/Tris citrate pH 8.3 jel ve elektrot sistemine göre elektroforezden geçirilip Wendel ve Weeden (1989) yöntemine göre boyandığı kombinasyon en iyi sonucu vermiştir.
Determination of the most effective method for peroxidase isozyme analysis by starch gel electrophoresis in pear and quince
This study was conducted to determine the most effective starch gel electrophoresis (SGE) method for peroxidase (PRX) analysis that can be used various aimes in pear varieties and quince rootstock. Bark tissues collected from current year shoots of Bartlet (ET-Williams) and Beurre Hardy (EH) pear scions and QA rootstock were used in this study. In SGE, 3 different extraction buffers, Krebs (1996), modified Krebs (1996) and modified Santamour et al (1986); 3 different gel and electrod buffers, Histidine-citrate pH 5.7, Morpholine-citrate pH 6.25 and Lithium Borate pH 8.3; and 3 diffenet staining buffers, Wendel and Weeden (1989), modified Santamour et al. (1986) and Santamour et al. (1986) Guaiacol method were used. As a result, the combination of method consisted of Santamour et al. (1986) extraction buffer, Lithium borate/Tris citrate pH 8.3 gel and electrod buffer and Wendel and Weeden (1989) satining buffer was given the best isozyme pattern for peroxidase analysis of pear and quince by SGE.
