Sialoadenektomi ve eksojen epidermal growth faktör uygulamasının sıçan uterus dokusu üzerindeki etkilerinin araştırılması

Sıçanlarda submandibular bez zengin bir Epidermal Growth Factor'ün (EGF) kaynağıdır. Çalışmanın amacı; sialoadenektomi operasyonu yapılarak, endojen EGF eksikliği oluşturulan sıçanlara dışardan verilen EGF'nin uterus dokusu üzerindeki etkilerinin histolojik olarak araştırılmasıdır. Bu amaçla; erişkin 30 adet 180-200 günlük ve 220-250 gr ağırlığında Wistar Albino türü sıçanlar rastgele 3 gruba ayrıldılar. Grup-I: Kontrol(n= 10), Grup-II: Sialoadenektomi(n=10),Grup-III: Sialoadenektomi operasyonundan 22 gün sonra 75 mikrogram/5 gün sure ile ekzojen EGF (n=10). Hayvanlar çalışmanın başlangıcından 27 gün sonra pentobarbital ile sakrifiye edilerek, uterus dokuları alındı. Alınan doku örnekleri bouin ve %10'luk nötral formalinde fikse edildi. Rutin histolojik işlemlerden sonra, elde edilen kesitler Hematoksilen-Eosin, Van Giesson ve Mallory Azan ile boyanarak ışık mikroskobunda değerlendirildi. Sialoadenektomi işlemine bağlı olarak; uterus epitelinde, endometrial bağdokusunda ve uterinal bezlerde histolojik değişiklikler izlendi. Sialoadenektomi sonrası dışarıdan verilen EGF'nin, histolojik değişiklikleri kısmi olarak düzelttiğini gözlemledik. Sonuç olarak; tükürük bezin'den salgılanan EGF'nin uterus dokusu üzerinde etkili olabileceği kamsına vardık.

The effects of exogenous epidermal growth factor administration in sialoadenectomy rats on the uterus tissue: A light microscopic study

The submandibular gland is a rich source of epidermal growth factor Epidermal Growth Factor (EGF) in rat. The aim of this study was to determined the histological effects on the uterus of the sialoadenectomized (surgical removal of the submandibular glands) rats which were occured endogenous EGF deficiency followed by administered exogenous EGF. Thirty adult female Wistar-Albino rats weighing between 220 and 250 g were randomly divided into three groups. Group-I; Control (n= 10), Grup-II; Sialoadenectomized rats (n=10), Group-Ill; 22 days after sialoadenectomy, exogenous EGF (75 microgram, Sigma E9644) (n=10) was added to the tapwater for 5 days, Twentyseven days after the study was started, animals were sacrified by sodium pentobarbital 50 mg/kg, IP and uterinal tissue samples was fixed in 10% neutral formalin and Bouin's solutions. After histological procedures were carried out, sections were stained with Haematoxylin-Eosin, Van Giesson, and Mallory Azan and monitored on the light microscope. Histological changes related to the sialoadenectomy were observed in the epithelium, connective tissue and endometrial glands of the uterus. After sialoadenectomy, addition of the exogenous EGF to the water was partially recovered histological changes on the uterinal tissue. Finally, the section of EGF from the submandibular gland plays an important role on the uterinal tissue.
