Dişi köpeklerde östrus sırasında vaginal sitolojiye dayalı asidofilik indeks (Aİ) ve serum progesteron seviyeleri üzerinde çalışmalar

Çalışmada pubertaya ulaşmış yedi adet melez dişi köpek kullanıldı. Köpeklerden proöstrus belirtilerinin başlamasından diöstrusun ilk gününe kadar hergün kan ve vaginal smear örnekleri toplanarak incelendi. Kan örneklerinde progesteron seviyesinin 5.0 ng/ml'ye ulaşması ovulasyon zamanı (0. Gün) olarak kabul edildi. Bundan 5 gün önce ve sonrasında alınan kan ve smear örnekleri değerlendirildi. Kan serumundaki progesteron hormon konsantrasyonları RIA yöntemi ile belirlendi. Pamuklu çubuk tekniği kullanılarak alınan vaginal smear örnekleri Pappanicolaou boyama tekniği ile boyandı ve incelenen örneklerde asidofilik index (Aİ) saptandı.Çalışmada en yüksek Aİ %70±3.8 ile ovulasyondan sonra 1. günde saptandı. Aİ 3. günde (%40±7,7) hızla düştü ve bu düşme 5. güne kadar sürdü. En yüksek Aİ değerleri ovulasyondan bir gün önce, ovulasyon anında ve ovulasyondan bir gün sonra saptandı. Serum progesteron seviyeleri ise ovulasyondan 5 gün önce 0.7±0.2 olarak bulundu ve giderek artarak ovulasyondan 5 gün sonra 16±4.2 ng/ml'ye ulaştı. . Dişi köpeklerde pamuklu çubuk yöntemi ile alman vaginal smear örneklerinde saptanan asidofılik indeksin, ovulasyon zamanını tahmin etmede yararlı olduğu sonucuna varıldı.

Determination of ovulation time using the eosinophilic index (EI) of vaginal cytology and serum progesterone levels in the bitch

In this study, cross-bred, pubertal, seven bitches were used. The bitches were bled every day and vaginal smears were taken after the sign of proestrus until the first day of diestrus. The ovulation time (day Q) was accepted as the progesterone level in sera reached 5 ng/ml. The smears and sera taken 5 days before and 5 days after the ovulation time were evaluated. The concentration of progesterone in blood sera were measured by radioimmunoassay (RIA). The smears on cotton-swabs were stained with Pappanicolaou stain and the eosinophilic index (El) were determined. The results of this study showed that the highest El (70±3.8%) was determined a day after the ovulation time. The El value decreased dramatically at day 3 (40±7,7%) and the decrease lasted by day 5. The highest El values were determined a day before the ovulation, at ovulation and a day after the ovulation. The progesterone level in sera was 0.7±0.2 ng/ml 5 days before the ovulation and increased by day 5 after the ovulation and reached at 16±4.2 ng/ml. This study has indicated that the El value obtained from the cotton-swabs smears should be considered for prediction of ovulation in bitches.
