TOKAT VE NEVŞEHİR YÖRELERİNDE YETİŞEN NARİNCE ÜZÜMÜ VE ÜZÜM ŞIRASININ SPONTAN FERMANTASYONU SIRASINDA MAYA POPÜLASYONUNUN PZR–RFLP TEKNİKLERİ İLE BELİRLENMESİ
Bu çalışmada, 2015 yılında Tokat ve Nevşehir yörelerinden bağbozumu dönemlerinde hasat edilenNarince üzümlerinin spontan fermantasyonu sırasında ortamda bulunan S. cerevisiae ve Saccharomycesspp. olmayan mayaların popülasyonu incelenmiştir. Mayaların moleküler yöntemle tanımlanmasındaPZR–RFLP yöntemi kullanılmıştır. PZR yönteminde mayalar öncelikle 5.8S rRNA’larında bulunan ITSgen bölgelerinin boyutlarına göre gruplandırılmış, daha sonra 26S rRNA’larına ait D1/D2 gen bölgeleriile yapılan dizi analizi sonuçlarıyla mayalar tür düzeyinde tanımlanmışlardır. Narince üzümlerinden veüzüm şırasının spontan fermantasyonu sırasında ortamdan 189 adet maya izole edilmiştir. Tokat yöresihasat üzümlerinden yalnızca H’spora uvarum, Nevşehir yöresi hasat üzümlerinden ise yalnızca S.cerevisae izole edilmiştir. Narince şırasının spontan fermantasyonu sırasında ise H’spora uvarum,H’spora guilliermondii, S. cerevisiae, T. delbrueckii, W. anomalus, P. kluyveri, I. terricola, I. orientalis,P. occidentalis, L. thermotolerans, Metchinikowia spp. ve C. zemplinina, türleri tanımlanmıştır.
DETERMINATION OF YEAST POPULATION BY PCR–RFLP TECHNIQUES ISOLATED FROM SPONTANEOUS FERMENTATION AND NARİNCE GRAPES GROWN IN TOKAT AND NEVŞEHİR
In this study S. cerevisiae and non–Saccharomyces populations were investigated during spontaneous alcoholic fermentation of Narince grapes grown in Tokat and Nevşehir harvested in 2015 vintage. Yeasts were identified by PCR–RFLP analysis of the 5.8 ITS rRNA region and sequence information for the D1/D2 domains of the 26S gene. During spontaneous fermentation of Narince, 189 yeasts were isolated. H’spora uvarum was only yeast isolated from Narince grapes obtained from Tokat region, however only S. cerevisiae was isolated from Cv. Narince harvested from Nevşehir region. During spontaneous fermentation of Narince grapes H’spora uvarum, H’spora guilliermondii, S. cerevisiae, T. delbrueckii, W. anomalus, P. kluyveri, I. terricola, I. orientalis, P. occidentalis, L. thermotolerans, Metchinikowia spp. and C. zemplinina species were identified.
