Effekte von phenylhydrazin toxizitat und silymarin über die synthese von MT-I/11 bei syrischen hamstern
Bu çalışmada deneysel olarak oluşturulan Fenilhidrazin (PH) toksikasyonunda Silimarinin (S) rolü ve gruplar arasında Metallothionein (MT) sentezindeki farklılıkların immünohistokimyasal yöntemle ortaya konulması amaçlandı. Çalışmada 40 adet Suriye Hamsteri kullanıldı ve hayvanlar 4 eşit gruba (kontrol grup, PH grup, Silimarin, Phenylhydrazin+Silymarin [PHS] grup) ayrıldı. Kontrol grubu hayvanlarına intraperitoneal yolla %0,9 NaCl, PH grubuna intraperitoneal yolla fenilhidrazin, PHS grubuna ise intraperitoneal yolla fenilhidrazin ve oral yolla silimarin 10 gün süreyle uygulandı. Silimarin gurubuna oral olarak silimarin 10 gün süreyle verildi. Çalışmada uygulamaların bitimini takiben hayvanlar servikal dislokasyonla öldürüldü. PH toksisitesine bağlı olarak karaciğer, dalak ve böbrekte makroskopik ve mikroskopik değişikliklere rastlandı. Silimarinin PH’nin neden olduğu toksisiteye bağlı olarak ortaya çıkan doku değişikliklerini azalttığı görüldü. PH’nin kontrol grubuna göre MT sentezinde bir artışa neden olduğu ortaya konuldu. PHS grubunda yalnızca PH uygulanan gruba göre daha düşük oranda MT-sentezinin gerçekleştiği gözlendi. Sonuç olarak oral yolla verilen Silimarinin, PH toksisitesine karşı hücresel savunma mekanizmasında etkili olabileceği sonucuna varıldı.
Suriye hamsterlerinde fenylhidrazin toksisitesi ve silimarinin MT-1/11 sentezi üzerine etkileri
Das Ziel der vorliegenden Studie war die Rolle des Silymarins bei der experimental Phenylhydrazin (PH)-Toxizität und die Unterschiede von Metallothionein (MT)-Expression mit Hilfe der immunohistochemischen Techniken zwischen den Gruppen zu untersuchen. In dieser Untersuchung dienten insgesamt 40 syrische Hamster. Die Tiere wurden in vier Gruppen als Kontroll- und drei Untersuchungsgruppen (PH, Phenylhydrazin+Silymarin [PHS], Silymarin Gruppe) aufgeteilt. Die bei den Hamstern durchgeführten Behandlungen dauerten 10 Tagen. Die Kontrollgruppe erhielt intraperitoneale Injektionen von 0,9% NaCl. Die PH-Gruppe erhielt tägliche die intraperitonealen Injektionen von Phenylhydrazin und die PHS-Gruppe intraperitoneale Injektionen von Phenylhydrazin und orale Gabe von Silymarin. Der Silymaringruppe (S-Gruppe) erhielt die intraperitoneale Injektionen von 0,9% NaCl und orale Gabe von Siliymarin für 10 Tage. Am Ende der Durchführung wurden allen Hamster mit Zervikal dislokation getötet. Es konnte festgestellt, dass die intraperitoneale Phenylhydrazinapplikationen die makroskopische und mikroskopische deutliche Veränderungen in der Leber und Milz sowie in den Nieren bei Hamster verursachen. Es wurde gezeigt, dass die Silymarin Gabe die durch PH-Tokzität verursachten Veränderungen in den Organen vermindert und dass PH zu einer Erhöhung der MT-Expression bei der PH-Gruppe im Vergleich zu der Kontrollgruppe und PHSGruppe. Zusammenfassend, dass intraperitonale Injektionen von Phenylhydrazin (PH) ausgeprägte Veränderungen bei Hamster verursachen und dass Silymarin eine Rolle in den zellulären Abwehrmechanismen spielen kann.
