Effect of serum gonadotrophin (PMSG) administration on oestrus and ovulation in anestrous bitches
Bu çalışmada köpeklerde östrus ve ovulasyonun uyarılmasında gonadotropin uygu-lamalarının etkisi araştırılmıştır. Çalışmada 8 Husky ve 7 Collie ırkı fertilité problemi olan köpekler kullanılmıştır. Gonadotropin uygulamalarından bir gün önce alman vaginal smear örneklerine göre bir köpeğin diöstrus, diğerlerinin anöstrusta oldukları saptandı. I.grup hayvan-lara günlük 44 LU. PMSG/kg ile 9 gün gonadotropin uygulamasına başlandı. Uygulamalar sırasında proöstrus kanaması başlayan köpeklerde gonadotropin enjeksiyonlarına son verildi. Vaginal smear ile östrusa girdikleri tesbit edilen köpeklere östrusun l ve 2. Günleri 500 I.U. hCG uygulanarak tohumlandılar. Il.grup köpeklere 6 gün ara ile iki kez günlük 500 LU. PMSG ve östrusun l ve 2. Günleri 500 LU. hCG uygulandı. III. Grup köpeklere uygulama yapılmadan kontrol olarak bırakıldılar.Çalışma süresince günaşırı kan ve vaginal smear örnekleri alınarak oluşturulan östrus sik-lusu takip edildi. II ve III. grup köpeklerde hormonal, hücresel ve davranışsal değişim gözlenmezken, I.grup köpeklerde östrus ve ovulasyon uyarılabilmiştir. I.grup köpeklerde gonadotropin uygulamasının 3. günü seram progesteron ve östradiol seviyelerinde artış sağlanmıştır. Proöstrus döneminde en yüksek seviyeye çıkan östradiol seviyesi östrusun başlamasıyla düşmüştür. Proöstrus sonunda artmaya başlayan serum progesteron seviyesi östrusta ovulasyonu takiben hızla artmaya devam etmiştir.Günlük 44 IU PMSG uygulaması ile anöstrus köpeklerde östrus ve ovulasyonlar uyarılabilmiş ve 7 günlük uygulama yeterli bulunurken 6 gün ara ile 2 kez 500 IU PMSG uygulaması anöstrus köpeklerde östrus ve ovulasyonun uyarılmasında etkili bulunmamıştır.
Anostrus köpeklerde serum gonadotropin (PMSG) uygulamalarının östrus ve ovulasyon üzerine etkileri
Summary: The effects of exogenous gonadotrophin treatments to induce oestrus and ovulation in bitches were investigated in this study. 8 Husky and 7 Collie infertile bitches wereused as material. One day before exogenous gonadotrophin treatments vaginal smears were examined and one bitch was diagnosed at diestrous, the rest were at anestrous. Animals in the first group were treated with daily 44 I.U.PMSG/kg for 9 days treatment program. PMSG injec-tions were stopped in the bitches which had proestrous bleeding. 2 bitches on the 3rd day and 4 bitches on the 7th day of treatment had proestrous bleeding. Bitches detected on heat by vaginal smear were injected with 500 I.U.hCH on the first and 2nd days of oestrus and inseminated. Bitches in the 2nd group received 2 injections of 5001.U.PMSG with 6 days interval and 500 I.U. hCG on first and 2nd days of estrous. The 3rd group had no treatment and considered as control. Blood and vaginal smear samples were collected every other days during the study and induced oestrus cycles were observed. No hormonal, cytological and behavioural differences were observed in 2nd and 3rd group bitches, while the first group bitches had induced oestrus and ovulation. Serum progesterone and oestradiol levels rose on the 3rd day of gonadothropin treat-ments. Oestradiol level which was the highest during proestrous decreased with the onset of estrous. Serum progesterone level began to increase at late proestrous and continued to rise dur-ing estrous after ovulation.Oestrus and ovulation could be induced with daily repeated 44 UI PMSG/kg injections and for 7 days treatment was sufficient although 2 injections of 500 IU PMSG with 6 days inter-val was not effective to induce oestrus and ovulation in anestrous bitches.
