Anaerobik Rumen Fungus İzolelerinin Rastgele Çoğaltılmış Polimorfik DNA (RAPD) Metodu ile Filogenetik Analizi

Bu çalışmada farklı cinslere ait anerobik rumen fungus izolatları arasındaki ilişkinin ortaya konması amacıyla filogenetik analiz, yapılmıştır. Türkiye'nin farklı coğrafi bölgelerinden izole edilen onsekiz fungus kültürü Rastgele Çoğaltılmış Polimorfik DNA (RAPD) tekniği ile analiz edilmiştir. İzole edilen onsekiz fungus kültürüne 10 baz uzunluğundaki primerler kullanılarak Polimeraz Zincir Reaksiyonu (PZR) uygulanmıştır. Toplam olarak 64 markır bandı gözlemlenmiş ve bu bantların %92.18'inin polimorfik olduğu saptanmıştır. Çoğaltılan DNA büyüklükleri 100 bç ile 700 bç arasında değişmektedir. Fungusları birbirinden ayrıt etmek için önemli derecede polimorfizim gözlemlenmiş ve izole edilen 18 fungusun tamamı açık bir şekilde birbirinden ayrılmıştır

Phylogenetic Analysis of Anaerobic Fungal Isolates by Randomly Amplified Polymorphic DNA (RAPD)

Phylogenetic analysis was conducted to asses the relationships between the isolates from the various genera of anaerobic rumen fungi. A set of 18 fungal cultures isolated and purified from the ruminant herbivores from different geographic locations of Turkey was analyzed with the aid of randomly amplified polymorphic DNA (RAPD) technique. Polymerase chain reaction (PCR) using 10 arbitrary decamer oligonucleotide primers applied to the 18 anaerobic fungal isolates. A Total of 64 different marker bands were observed and 92.18% of these bands were found to be polymorphic. The sizes of the amplified DNAs were mainly range between 100 bp and 700 bp. A remarkable polymorphism was detected to allow the separation of individuals and all the 18 fungal isolates were clearly distinguished from each other
