Üç aylık bir kuzuda kistik ekinokokkozis’in moleküler tanısı

Bu makalede Bolu’nun Kıbrısçık İlçesinde üç aylık bir kuzuda Echinococcus granulosus sensu stricto (G1-G3 grubu) enfeksiyonu rapor edilmiştir. Laboratuvarımıza gönderilen ölü bir kuzuya ait karaciğer, makroskobik olarak incelenmiş ve üzerinde biri 1.5 X 1.2 mm, diğeri 3.6 X 1.6 mm boyutlarında beyaz renkli iki odak tespit edilmiştir. Bu iki odak disekte edilerek karaciğerden ayrılmış ve DNA ekstraksiyonu yapılmıştır. Elde edilen genomik DNA’nın mitokondrial 12S rRNA geninin 254 baz çiftlik (bç) bölgesi PZR ile çoğaltılmış ve kistin E.granulosus s.s. kaynaklı olduğu belirlenmiştir. Çalışma sonucunda çok küçük çaplı kistlerin bile E.granulosus’un hangi grubu içerisinde yer aldığının PZR ile belirlenebileceği ortaya konulmuştur.

Molecular detection of cystic echinococcosis in a three month old lamb

In this article, Echinococcus granulosus sensu stricto (G1-G3 cluster) infection was reported in a three-month-old lamb in Kıbrısçık county of Bolu. Liver of the dead lamb which has been sent to our laboratory was macroscopically examined, and two foci with white color were diagnosed in sizes of 1.5 X 1.2 mm and 3.6 X 1.6 mm, respectively. These two foci were dissected from liver, and DNA extraction was performed. 254 bp region of mitochondrial 12S rRNA gene from acquired genomic DNA was amplified, and source of cyst was detected as E. granulosus s.s. As a result, we reveal that cluster of E.granulosus can be determined even in very small cysts by PCR.
