Antalya Yöresindeki Keçilerde Hemotropik Mycoplasma spp.’nin Nested PCR ile Araştırılması
Bu çalışma, hemotropik Mycoplasma spp.’nin Antalya yöresindeki keçilerde varlığı ve yaygınlığınınnested PCR ile ortaya konması amacıyla yapılmıştır. Bu amaçla Antalya ilinde bulunan 3 odaktaki(Akseki, Manavgat ve Serik) keçilerden toplam 157 adet kan örneği toplanmıştır. Çalışmadakullanılan hayvanlar ayrıca kene varlığı yönünden muayene edilmiştir. Bu örneklerden totalgenomik DNA’lar izole edilmiş, elde edilen DNA örnekleri hemotropik Mycoplasma spp. varlığıyönünden 16S rRNA gen bölgeleri nested PCR ile amplifiye edilmiştir. İncelenen 157 örneğin35’inde (%22.3) Mycoplasma spp. pozitifliği tespit edilmiştir. Pozitif örnekler arasından seçilen birörneğin 16S rRNA genine ait sekans analizi sonucu Mycoplasma ovis ile %100 benzer olduğugörülmüştür. Bu çalışma ile Antalya yöresindeki keçilerde nested PCR ile hemotropik Mycoplasmaspp. varlığı ve etkene ait epidemiyolojik veriler elde edilmiştir.
Investigation of Hemotropic Mycoplasma spp. by Nested PCR in the Goats in Antalya Region
This study was carried out in order to reveal the presence and prevalence of hemotropic Mycoplasma spp. in goats in Antalya region by nested PCR. For this purpose, a total of 157 blood samples were collected from the 3 focus of the Antalya province (Akseki, Manavgat and Serik). The animals used in the study were also examined for presence of ticks. Total genomic DNAs were isolated from these samples, and DNA samples were obtained from hemotropic Mycoplasma spp. the presence of the 16S rRNA gene region was amplified by nested PCR. 157 of the samples examined, 35 (22.3%) were Mycoplasma spp. positivity was detected. Sequence analysis of the selected among the positive samples, was found to be 100% similar to the result of Mycoplasma ovis. In this study, nested PCR of hemotropic Mycoplasma spp. presence and epidemiological data of the agent were obtained.
