Gentamisine duyarlı Staphylococcus aureus suşlarında gentamisinin biyofilm ve koagülaz oluşumuna etkisi
Klinikte önemli patolojilere neden olabilen Staphylococcus aureus’un patojenitesinde biyofilm ve plazma koagülaz enzimi oluşturmanın da önemi vardır. Bu çalışmada kateter ucundan ayrıştırılmış bir klinik suş (KSA) ile standart bir ATCC 25923 suşunun (SSA), gentamisinin en düşük baskılayıcı yoğunluk (MİK) altındaki ve üstündeki yoğunluklarda biyofilm ve bağlı ve serbest koagülaz oluşturma özellikleri karşılaştırılmıştır. Mikrodilüsyon yöntemi ile gentamisinin MİK değerleri KSA suşu için 4 µg/mL, SSA suşu için MİK 0.125 µg/mL olarak belirlenmiştir. Kristal viyole yöntemi ile belirlenen biyofilm oluşumunun ve lamda belirlenen bağlı koagülaz testinin iki suş için de MİK ve altındaki gentamisin yoğunluklarında inhibe edilmediği, MİK üstündeki yoğunluklarda ise inhibe edildiği saptanmıştır. Tüpte serbest koagülaz testinde ise ilk 2 saatte iki suşta da ancak belirli MİK altındaki yoğunluklarda inhibisyon görülürken, 4 ve 24 saatte KSA suşunda 2xMİK’de bile inhibisyon olmamıştır. Bu sonuçlara göre; her iki suşta da biyofilm oluşumunu başlatan genetik ekspresyonun, gentamisinin MİK yoğunluğunda bile pozitif bulunmasının klinikteki uygulamalarda dikkat edilmesi gereken bir özellik olduğu düşünülmüştür.
The effect of gentamicin on biofilm formation and coagulase activity in gentamicin susceptible Staphylococcus aureus strains
Staphylococcus aureus is an important pathogen which can cause severe pathologies in clinical practice. The forma-tions of biofilm and coagulase enzyme are important factors of its pathogenesis. In this study, biofilm formation and coagu-lase activity of a clinical strain (KSA) isolated from catheter and standart ATCC 25923 strain (SSA) were investigated in above and below minimum inhibitor concentrations (MIC) of gentamicin. The MIC values of gentamicin by microdilution method were found to be 4 µg/ mL for KSA strain and 0.125 µg/ mL for SSA strain. Biofilm formations detected by crystal violet method and slide (bound) coagulase tests were not inhibited in both strains by MIC and sub-MIC gentamicin concentrations. On the other hand, at concentrations above the MIC values these two assays were inhibited. Tube (free) coagulase assay was inhibited at certain sub-MIC concentration at the first two hours, but it was not inhibited at 4 and 24 hours in even 2xMIC concentration. As a result, the genetic expression of biofilm formation even at MIC concentrations seems to be important in clinical practice.
