Teke spermasının morfolojik değerlendirilmesinde farklı boyama metotlarının kullanılması

Bu çalışmada Saanen ırkı teke spermasının değerlendirilmesinde Eozin-nigrozin (EN), Gimza ve ikili boyama yöntemi olan Eozin-nigrozin sonrası Gimza (EN-G) boyama metotlarının kullanılabilirliği araştırıldı. EN, Gimza ve EN-G boyama yöntemlerinde akrozomal bozukluk (AB) oranları sırasıyla; %3.5±0.4, %7.2±0.4 ve %9.1±0.4; Diğer Morfolojik Bozukluk (DMB) oranı 2.8+0.2, 2.4+0.2 ve 2.8±0.2, Toplam Morfolojik Bozukluk (TMB) oranı ise sırasıyla 6.3+0.4, 9.6+0.4 ve 11.9±0.4 olarak saptandı. Evaluation of goat semen morphology after different staining methods

This study was designed to assess effectiveness of different staining methods, Eosin-nigrosin (EN), Giemsa and double staining method Eosin-nigrosin combined with Giemsa (EN-G), to detect dead spermatozoa, defected acrosome rates (AB), Other Morphological Defect (DMB) and Total Morphological Defect (TMB) in fresh Saanen semen. The percentages of spermatozoa with AB, DMB and TMB were 3.5%, 7.2% and 9.1%; 2.8%, 2.4% and 2.8%; and 6.3%, 9.6% and 11.9% in EN, Giemsa and EN-G, respectively. The highest AB was observed in EN-G staining method (P<0.05). There were no significant differences in percentage of dead spermatozoa between EN and EN-G staining methods. In conclusion, EN-G staining method could be used to distinguish the percentage of dead spermatozoa, AB, DMB and TMB in the same slide.
