Arcobacter türlerinin atık kuzulardan izolasyonu ve multipleks PCR ile identifikasyonu
Arkobakterler aerobik ortamda ve daha düşük ısılarda üreyebilmeleri bakımından kampilobakterlerden ayırılırlar. Bu cins, patojen olan Arcobacter butzleri, A. cryaerophilus ve A. skirrowii türlerini içermekte ve bu mikroorganizmalar insan ve hayvanlarda abort, enterit ve mastit gibi bir çok hastalıkla ilişkilendirilmektedir. Bu çalışma, Arcobacter türlerini atık kuzulardan izole etmeyi ve izolatları multipleks PCR ile tür düzeyinde identifıye etmeyi amaçlamıştır. Çalışma materyalini, 2005-2006 kuzulama döneminde Kars bölgesinden temin edilen 32 atık kuzu örneği oluşturdu. Bakteriyolojik ekim amacıyla fötal akciğer ve abomazum içeriği kullanıldı. Bir adet öme.K. Arcobacter-benzeri organizma yönünden pozitif bulundu ve bu izolat multipleks PCR tekniği ile A. cryaerophilus olarak identifıye edildi. Bu sonuçlar, A. cryaerophilus ve diğer Arcobacter türlerinin koyun atıkları etiyolojisindeki önemini vurgulamaktadır. Aynı zamanda, koyun abortlarına karşı alınması gereken koruma ve kontrol yöntemlerinde Arcobacter türlerinin de dikkate alınması gerekliliğini ortaya koymaktadır.
Isolation of arcobacter spp. from aborted sheep fetuses and identification of the isolates by multiplex PCR
Arcobacters are differentiated from campylobacters by their ability to grow under aerobic condition and at lower temperatures. This genus includes three pathogenic species, Arcobacter butzleri, A. cryaerophilus and A. skirrowii, increasingly found to be associated with a variety of human and animal diseases such as abortion, enteritis and mastitis. The present study aimed to isolate Arcobacter spp. from aborted sheep fetuses and to identify the isolates at the species level by multiplex PCR. Thirty two aborted sheep fetuses were used to isolate Arcobacter spp. during 2005-2006 lambing season in Kars region. Fetal lung and stomach contents were used for bacteriological inoculations. Arcobacter-like organisms were found in one sample and multiplex PCR analyses of this isolate identified the organism as Arcobacter cryaerophilus. This suggests that A. cryaerophilus and other Arcobacter spp. should be taken into account during examining sheep abortions. Arcobacter spp. should be considered for applying appropriate measures in order to control and prevent abortions.
