Sığır oositlerinde morfolojik görünümün in vitro olgunlaştırma ve fertilizasyon üzerine etkileri
Morfolojik görünümün in vitro olgunlaştırma ve fertilizasyona etkisinin araştırıldığı bu çalışmada, mezbahada kesilen inek ovaryumlarından elde edilen oositler 3 ayrı aşamada (1: kazanıldıktan hemen sonra; 2: in vitro olgunlaşma sonrası; 3: in vitro fertilizasyon sonrası) kullanıldı. Herbir aşama için, elde edilen oositler stereo mikroskop altında değerlendirilerek 3'er gruba ayrıldı. Grup 1: En az 4 sıra kompakt kumulus hücre kitlesi bulunan ve homojen vitellusa sahip oositler; Grup 2: 1-4 sıra kumulus hücre kitlesi bulunan oositler, ve Grup 3: Üzerlerinde kumulus hücresi bulunmayan, ekspansi-yona uğramış veya kumulus hücreleri dejenere olmuş oositler. Birinci aşamada (kazanmadan hemen sonra) incelenen 19, 33 ve 28 oositin sırasıyla 0, 2 (%6.1) ve 2 (%71) tanesinin mayoza başlamış, 1 (%5.3), 2 (%6.1) ve 3 (%10.7) tanesinin ise dejenere olduğu, yani in vitro olgunlaştırma ve fertilizasyon amacıyla kullanılamayacağı tespit edildi. Gruplar arasındaki farklar istatistiki olarak an¬lamlı bulunmadı (p>0.05). İkinci aşamada (in vitro olgunlaştırmayı takiben), morfolojik gruplara göre incelenen 28, 33 ve 12 oositten sırasıyla 16 (%57.1), 14 (%42.4) ve 7 (%58.3) tanesinin metafaz II (M II) dönemine ulaştığı saptandı. Gruplar arasındaki fark¬lar istatistiki olarak anlamlı bulunmadı (p>0.05). The effects of morphological appearences on the In vitro maturation and fertilization of bovine Oocytes
In this study during which the effect of morphological appearance on in vitro maturation and fertilization was investigated, oocytes collected from the ovaries of slaughtered cows were used in 3 different steps (1: immediately after collection; 2: after in vitro maturation; 3; after in vitro fertilization). In every step, oocytes were evalu¬ated under a stereo microscope and divided into 3 other groups. Group 1: Oocytes hav¬ing at least 4 layers of compact cumulus cells and a homogenous vitellus; Group 2: Oo¬cytes having 1-4 layers of compact cumulus cells; Group 3: Oocytes not having or hav¬ing degenerated cumulus cells, or expanded oocytes. In the first step (immediately after collection), out of 19, 33 and 28 oocytes 0, 2 (6.1%) and 2 (7.1%) were observed to resume meiosis; 1 (5.3%), 2 (6.1%) and 3 (10.7%) were observed to be degenerated, respectively, and not suitable for in vitro, maturation and fertilization. The differences between groups were not significant statistically (p>0.05). In the second step (after in vitro maturation), out of 28, 33 and 12 oocytes which were examined according to the groups, 16 (57.1%), 14 (42.4%) and 7 (58.3%) were observed to reach metaphase II (M II) stage, respectively.
