Agrotis segetum (Denis ve Schiff.) (Lepidoptera: Noctuidae) hemolenf lektinin hemaglutinasyonu

Agrotis segetum'un 5. evre larvalarından alınan hemolenf lektininin, in vitroda çeşitli omurgalı eritrositlerini (horoz, koyun, tavşan, sığır ve A,B,0 grubu insan kanı) hemaglutinasyonu incelenmiştir. Deneylerde kullanılan hemolenf lektin örneklerinin en yüksek titre ile (1:256), A grubu insan kanı eritrositlerini, en düşük titre ile (1:4) at ve sığır kanı eritrositlerim hemaglutine ettiği tesbit edilmiştir. Çalışmada bazı karbohidratlann ve glikoproteinlerin lektin hemaglutinasyonu aktivitesininin inhibisyonu testleri de yapılmıştır. Hemolenf lektini hemaglutinasyon aktivitesini N-asetilgalaktozamin, N-asetilglukozamin , D-fruktoz, D-galaktoz, D-glukoz, D-mannoz, D-rafinoz, L-riboz gibi deneylerde kullanılan bazı karbohidratlar inhibe etti. Fakat D-laktoz ve mucin'in (Img/mL) (glikoprotein) inhibisyonu diğerlerine göre daha etkili olmuştur. Öte yandan, bütün karbohidratlar O grubu insan kam hemaglutinasyoununu inhibe etmemiştir.Hemaglutinasyon örnekleri (O grubu insan kanı eritrositlerine karşı) geçirmeli elektron mikroskobunda (TEM) incelenmiştir. Pozitif sonuç alınan hemaglutinasyon örneklerinde hemolenf lektininin eritrositleri birbirine bağladığı gözlenmiştir. Buna karşılık, negatif sonuç alınan örneklerde ise eritrositlerin bağlanmadığı gözlenmiştir.

Hemagglutination of hemolymph lectin agrotis segetum (Denis and Schiff.) (Lepidoptera: Noctuidae)

We examined that in vitro hemagglutination of hemolymph lectin samples, obtained from fifth instar larvae of Agrotis segetum was examined on several vertebrate (chicken, sheep, rabbit, bovine and human ABO blood) erythrocytes. Hemagglutination of used hemolymph lectin samples in experiments was determined highest titre (1:256 ), on human blood groupA and lowest titer (1:4) on horse and bovine erythrocytes.In the study, inhibition tests of hemagglutination activitiy were done by some carbohydrates and glycoproteins. Hemagglutination activity of hemolymph lectin were inhibited by some carbohydrates used in experiments such as N-acetylgalactoseamine, N-acetylglucoseamine, D-fructose, D-galactose, D-glucose, D-maltose, D-mannose, D-raffinose, L-ribose , but inhibition of D-lactose and mucin (1 mg/ml)(glycoprotein) were stronger than others. On the other hand, all carbohydrates were no inhibition hemagglutination of human blood O group. Samples of hemagglutination (against human O blood group) were examinedwith transmission electron microscope (TEM). Due to hemolymph lectin, each of erythrocytes was bound in positif showing result. Whereas, each of erythrocytes was not bound in negatif showing result.
