Gebe olmayan izole sığır uterus kontraksiyonları üzerine ceftiofur'un etkileri

Bu çal ı ş mada üçüncü ku ş ak bir sefalosporin olan ceftiofurun izole gebe olmayan inek ve düvemiyometriyumunun kendili ğ inden kas ı lmaları üzerine etkileri ara ş tı rı ld ı .Siklusun foliküler döneminde bulunan inek ve düvelerden al ı nan miyometriyal ş eritler, %95 oksijen%5 karbondioksit içeren gaz ile sürekli gazlanan, 38 ° Cdeki ve pHs ı 7.4 olan, 5 mL Krebssolüsyonu bulunan izole organ banyosuna as ı ld ı . Doksan dakikal ı k gerime uyum süresi sonunda,düzenli kendili ğ inden kas ı lma gösteren ş eritlerin kas ı lmaları , izometrik güç çevirgeci kullan ı larak 10dakika süre ile kaydedildi ve bu veriler kontrol olarak kullan ı ld ı . Daha sonra ş eritlerin bulundu ğ ubanyo s ı vı s ı na 500 µM, kümülatif 1000 µM ve 2000 µM dozları nda ceftiofur ilave edildi (n=16). Herdoz eklemesinden sonra kas ı lmalar 10 dakika süreyle kaydedildi. Ceftiofurun 10 dakikal ı kperiyotta, kendili ğ inden kas ı lmaları n s ı kl ı k, büyüklük ve e ğ ri altı nda kalan alan (EAKA)parametreleri üzerine etkisi de ğ erlendirildi. Effects of ceftiofur on conctractions of uterine isolated from non-pregnant cows

This study investigated the effects of ceftiofur, a third -generation cephalosporin on spontaneouscontractility of the myometrium isolated from non-pregnant cows and heifer uterus.Myometrial strips were isolated from cows and heifers that were at follicular phase and suspendedin a 5 mL jacketed organ bath filled with Krebs solution at 38 °C (pH 7.4) continuously bubbled with95% oxygen and 5% carbon dioxide. After manifestation of the spontaneous contractions duringequilibration period of 90 minutes, isometric contractions were recorded by using isometric forcedisplacement transducer and initial 10 minute was considered as control period. Then, the testsubstances ceftiofur (500 µM, cumulative 1000 µM and 2000 µM, n=16) were added to the tissuebath and evaluated by 10-min intervals. The effects of ceftiofur on amplitude (mg), frequency andarea under the contractile curve (AUC) of spontaneous contractions were evaluated by 10 minintervals. Data were statistically analyzed by using the Wilcoxon Signed Ranks Test and Pearson sCorrelation Test.It was determined that all tested doses of ceftiofur reduced the frequency of contractions (P0.05). After 500 µM and cumulative 1000 µM ceftiofur, theincrease in the AUC found to be statistically insignificant (P>0.05).
