Buzağıların Genişlemiş Spektrumlu Beta Laktamaz Üreten Escherichia coli Taşıyıcılığı
Gram negatif bakteriler tarafından üretilen genişlemiş spektrumlu beta laktamazlar (GSBL)bakterinin tüm penisilinlere, 1.-4. kuşak sefalosporinlere ve monobaktamlara dirençli olmasınısağlamaktadır. Bu çalışmada Türkiye’nin Burdur ili’ndeki sağlıklı buzağılarda GSBL üreten E. colitaşıyıcılığı ve bu izolatlarda yaygın GSBL genlerinin varlığı araştırıldı. Toplanan 150 buzağı dışkıörneğinin herbirinden tamponlanmış peptonlu su içinde %10’luk süspansiyon hazırlandı vesefotaksim ve seftazidim ilave edilerek hazırlanmış E. coli / koliform selektif agar besiyerlerine eşzamanlı ekildi. Besiyerinde görülen kolonilerden E. coli identifikasyonu standart metotlarla vepolimeraz zincir reaksiyonu (PZR) ile genotipik doğrulama yoluyla yapıldı. Ardından E. coliizolatlarının GSBL üretip üretmediği kombine disk metodu ile belirlendi ve yaygın GSBL genleriolan Sefotaksimaz-Münih (CTX-M), Temoneira (TEM) ve sülfhidril değişkeni (SHV) PZR ilearaştırıldı. Son olarak izolatların çeşitli beta laktam antibiyotiklere ve diğer sınıftan antibiyotiklereduyarlılığı agar disk difüzyon testi ile araştırıldı. Toplam 44 adet E. coli selektif agardan izole edildiancak bunların 36’sı GSBL üreten izolat olarak tespit edildi. CTX-M (grup 1) genleri tüm izolatlardabelirlenirken SHV genine hiçbir izolatta rastlanmadı. GSBL üreten E. coli izolatları beta laktamgrubu ve diğer grup antibiyotiklere yüksek oranda dirençli bulundu. Sonuç olarak, bu çalışma ileTürkiye’nin Burdur ilindeki işletmelerdeki sağlıklı buzağılarda GSBL üreten E. coli varlığı ve GSBLgen tiplerinin yaygınlığı gösterilmiş oldu.
Carriage of Extended Spectrum Beta Lactamase Producing Escherichia coli in Calves
Extended spectrum beta lactamases (ESBL) produced by Gram negative bacteria confer resistance to all penicillins, 1st-4 th generation cephalosporins and monobactams. In the present study, existence of ESBL producing E. coli isolates and frequency of the most prevalent GSBL genes in such E. coli isolates were investigated in the healthy calves in Burdur city of Turkey. A 10% suspension was prepared from each faeces (n=150) in peptone buffered water and spread on E. coli / coliform selective agar supplemented with cefotaxime and ceftazidime. The colonies appeared on the media were identified as E. coli by standard methods and genetic confirmation for E. coli was performed by polymerase chain reaction (PCR). Then ESBL production of E. coli isolates were confirmed by combined disc method and the common ESBL genes CefotaximaseMunich (CTX-M), Temoneira (TEM) and Sulfhydryl variable (SHV) were investigated by PCR. Finally, susceptibilities of the ESBL producing E. coli isolates to several beta lactams and other classes of antibiotics were investigated by agar disc diffusion test. Forty four E. coli isolates were isolated from selective media but 36 of them were confirmed as ESBL producer. CTX-M (group 1) genes were found in all isolates but SHV gene was not detected in the isolates. ESBL producing E. coli isolates were found resistant at high rate against to beta lactam group and other groups of antibiotics. Consequently, this study showed the presence of ESBL producing E. coli in the healthy calves in Burdur city of Turkey and also prevalence of types of ESBL genes in ESBL producing E. coli isolates.
