Bovine Ephemeral Fever Virüsün Rekombinant G Proteininin İmmunoperoksidaz Metodu ile Belirlenmesi

Bu çalışmanın amacı, bovine ephemeral fever virusun (BEFV) rekombinant glikoproteini (G) ile transfekte hücrelerin immunoperoksidaz (IP) yöntemi ile belirlenmesidir. Bu amaçla, BEFV G genini bulunduran rekombinantpcDNA4/HisMax(pcDNA4-G) vektörü Vero hücrelerine transfekte edilmiştir. Daha sonra, BEFV için spesifik monoklonal antikorlar kullanılarak gerçekleştirilen IP yöntemi ile G proteininin açıklatılmasının hücresel lokalizasyonu belirlenmiştir. Çalışmanın sonuçları IP yönteminin transfeksiyonun ve açıklatılmanın görüntülenmesi için ucuz, hızlı ve uygun bir yöntem olduğunu gösterdi.

Detection of Recombinant G Proteins ofBovine Ephemeral Fever Virus by Immunoperoxidase Method

The aim of this study was detection of the cells that transfected with recombinant glycoproteins (G) of bovine ephemeral fever virus (BEFV) by immunoperoxidase (IP) method. For this aim, the recombinant vector pcDNA4/HisMax (pcDNA4-G) including BEFV G gene was transfected into Vero cells. Immunoperoxidase (IP) method was used to investigate the intracellular localization of G protein expression using the spesific monoclonal antibody for BEFV G protein. According to our results, the IP method is affordable, rapid and suitable detection method for screening of transfections and expression
