Asmada (Vitis vinifera cv. Sultani Çekirdeksiz) Bax İnhibitor-1 Geninin İzolasyonu ve Moleküler Karakterizasyonu
P rogramlanmış hücre ölümü (PCD) canlılarda, gelişmede ve çevresel stres faktörlerinin etkisi ile zarar görmüş hücrelerin elimine edilmesinde önemli bir fizyolojik olaydır. Bax inhibitor-1 (BI-1) proteininin hücre ölümünü engellediği ve ökaryotlarda yüksek oranda korunduğu gösterilmiştir. Bu çalışmada, Vitis vinifera cv. Sultani Çekirdeksiz çeşidinin üzüm tanelerinden gen izole edilmiş ve VvBI-1 olarak adlandırılarak gen bankasına kayıt altına alınmıştır. Olası VvBI-1 geninin protein kodlayan kısmı 747 bç uzunluğundadır ve protein büyüklüğü de 248 aa olarak bulunmuştur. VvBI-1 protein dizisinin 27,290 kDa büyüklüğünde olduğu ve olası izoelektrik noktasının da 8,62 olduğu öngörülmüştür. Farklı gelişme dönemlerinde hasat edilmiş üzüm tanelerinden semi quantitative RT-PCR analizi ile VvBI-1 geninin ifade profili çıkarılmış ve VvBI-1 ifadesinin yüksek oranda ve tüm gelişme dönemlerinde var olduğu gözlemlenmiştir
