Koyunlara damar içi ve kas içi tek doz uygulanan uzun etkili oksitetrasiklinin biyoyararlanımı ve farmakokinetiği üzerine prednizolonun etkisi

Bu çalışmada, koyunlara tek doz (20 mg/kg, canlı ağırlık) kas içi ve damar içi uygulandıktan sonra uzun etkili oksitetrasiklinin farmakokinetiği ve prednizolon ile eş-zamanlı kas içi uygulamayı takiben de muhtemel farmakokinetik etkileşimin belirlenmesi amaçlandı. İlacın plazma yoğunluğu ELISA prosedürü ile ölçüldü. Kas içi ve damar içi uygulamadan sonra oksitetrasiklinin plazma yoğunluğu-zaman eğrisinin iki bölmeli açık modele uygunluğu tüm kinetik hesaplamalarda dikkate alındı. Oksitetrasiklin ve prednizolonun kas içi yolla kombine tarzda eş-zamanlı olarak uygulanması sonrası elde edilen farmakokinetik parametreler, tek başına uygulanan ilaç parametreleri ile benzerlik gösterdi (p>0.05). Dolayısıyla, sonuç olarak, uzun etkili oksitetrasiklinin tek başına veya prednizolon ile birlikte uygulandığında plazmada 48 saat boyunca ortalama terapötik düzeyi sağladığı ve prednizolonla birlikte uygulama durumunda ise olumlu veya olumsuz herhangi bir etkileşimin olmadığı kanısına varıldı.

The influence of prednisolon on the bioavailability and pharmacokinetics of long- acting oxytetracycline following intravenous and intramuscular administrations at a single dose in sheep

The purpose of this study was to determine the pharmacokinetics of long-acting oxytetracycline following intravenous and intramuscular administrations at a single dose (20 mg/kg body weight) and the likelihood of any interactions that might exist between the concurrent administrations of oxytetracycline and prednisolon by an intramuscular route in sheep. Plasma concentration of the drug was measured by ELISA procedure. Plasma oxytetracycline concentrations were fitted to a twocompartment open model. Following the intramuscular administrations of the long-acting oxytetracycline and prednisolon concurrently, the pharmacokinetic parameters obtained were similar to those with single administration without prednisolon (p>0.05). Therefore, it can be concluded that, both the administrations of long-acting oxytetracycline alone or together with prednisolon sustained the mean therapeutic plasma concentrations by 48 hours and there was no adverse or favourable interactions when the long-acting oxytetracycline and prednisolon were administered concurrently.
