Morchella esculanta Protein Hidrolizatlarında Anjiotensin Dönüştürücü Enzim I’in Peptid İnhibitörlerinin Araştırılması

Mantarlar sağlığa yararlı ve birçok dejeneratif hastalığa karşı koruyucu çok sayıda aktif bileşen içermelerinden dolayı alternatif ve tamamlayıcı tıpta terapötik ajanlar olarak kullanılmaktadır. Morchella esculenta dünyada yenilebilen en değerli mantar türlerinden biridir. M. esculenta mantarı antimikrobiyal, antiinflamatuvar, immunostimülatör, antitümör ve antioksidant gibi tıbbi ve farmakolojik özellikler sergileyen çok sayıda tokoferol, karotenoid, organik asit, polisakkarit ve fenololik bileşikleri içermektedir. Bu çalışmada M. esculenta mantarının angiotensin dönüştürücü I (ACE-I) enzimi üzerine inhibitör etkisi incelendi. Mantar ekstraktından alkalaz enzimi hidrolizi sonrası elde edilen peptid fragmanları ultrafiltrasyon membranları kullanılarak fraksiyonlandı. Elde edilen fraksiyonların angiotensin dönüştürücü I (ACE-I) enzimi üzerine inhibitör etkisi in vitro koşullarda çalışıldı. Fraksiyonlar içerisinde en yüksek ACE-I inhibisyonu % 94 ile 5kDa<peptidler<10 kDa arasındaki peptidleri içeren fraksiyonda gözlendi. Bu fraksiyonun ACE-I için IC50 değeri 4,2 µg/ml olarak hesaplandı. Fraksiyondaki peptid sayısı ve saflığı % 12 SDS-PAGE jel ile belirlendi. Çalışma sonuçları göstermektedir ki M. esculenta mantarı hipertansiyon hastalığının önlenmesi ve tedavisinde kullanılabilecek fonksiyonel peptid türevli gıda bileşenlerine sahiptir.

Investigation of Peptide Inhibitors of Angiotensin Converting Enzyme-I in Morchella esculenta Protein Hydrolysates

Mushrooms are use as therapeutics in alternative and complementary medicine as functional food because of contain a large number of biologically active components that offer health benefits and protection against many degenerative diseases. Morchella esculenta is one of the most highly priced edible mushrooms worldwide. It contains a wide range of active constituents which include tocopherols, carotenoids, organic acids, polysaccarides and phenolic compounds which exhibit a wide range of medicinal and pharmacological properties including anti-microbial, anti-inflammatory, immunustimulatory, antitumor and antioxidant. In this study; the peptide fragments were generated by alcalase hydrolysis from M. esculenta extracts were fractioned using ultrafiltration membranes. ACE inhibition were studied in the obtained fractions. The 5kDa <peptides< 10 kDa in the ultrafiltration fractions displayed highest ACE inhibition (94 %). The IC50 value of this fraction was calculated as 4,2 µg/ml. The peptide numbers and purity of fraction were determined with 12 % SDS-PAGE. The results indicate that M. esculenta derived peptides may have potential as functional food ingredients in the prevention and management of hypertension.
