Agresif ve kronik periodontitiste TNF-Ot ve lL-6 Gen polimorfizminin belirlenmesi

Investigation of TNF-Ot and lL-6 gene polymorphism in agressive and chronic periodontitis Background: Periodontitis is chronic inflammatory disease, and genetic factors may have an important role in its severity. Polymorphisms in the promoter regions of the lL-6 and TNF-oc gene have been reported to cause changes in the production of these cytokines. lL-6 polymorphisms have been shown to affect lL-6 promoter activity. The aim of this study was to evaluate the possible role of lL-6 174 and TNF-Ot 308 polymorphisms, in the severity of chronic and aggressive periodontitis. Methods: ln this study, group of 20 chronic periodontitis, 20 with severe chronic periodontitis and 20 healthy patients will be selected and will be performed in order to detect differences in genotype distributions of single nucleotide polymorphisms located in the promoter region of the lL-6 and TNF-Ot gene. DNA was isolated from all subjects using whole blood, and RT-PCR will be used to study the gene polymorphisms. Results: All clinical periodontal measurements were found higher both GKP and GAgP compared to controls (p>0.001), but there was no statistically significant differences between GKP and GAgP (p>0.05). When analyzed lL-6 174 G/C genotypes, there was no significantly differences between individuals and groups (p=0.017). All the groups of this study (GKP, GAgP and controls) were 100% G/G homozygous genotypes for TNF-Ot 308 G/A. There was no significantly differences among the groups (p=0.95). Conclusion: According of the results, there was no statistically relationship between the lL-6 174 and TNF 308 polymorphism and development of periodontal diseases.
