Rheum Ribes L. Ekstratını İçeren Sert Jelatin Kapsüllerin Kalite Kontrolünün Yapılması ve 1,1-Difenil 2-Pikril Hidrazil Serbest Radikal Süpürücü Aktivitesinin Değerlendirilmesi

Amaç: Rheum türleri tıbbi öneme sahiptir. Rheum ribes L. yerel olarak Işkın, Uçkun veya Uşgun olarak bilinmektedir. Genellikle Lübnan’da, İran’da ve Türkiye’nin doğusunda yetişmektedir. Rheum ribes L. bitkisinin çeşitli bölümlerinden elde edilmiş ekstraktlar birçok hastalığın tedavisinde kullanılmaktadır.Bu çalışmada, Anadolu folklorik tıbbında diyabet, hemoroid, ülser ve diyare tedavisinde kullanılan Rheum ribes L. bitkisinin ekstraktının hazırlanması, elde edilen toz ekstraktlarının Avrupa Farmakopesine uygun olarak toz kontrollerinin yapılması, hazırlanan ekstraktın sert jelatin kapsül formülasyonunun elde edilmesi ve Rheum ribes L. ektstraktını içeren sert jelatin kapsüllerin kapsül kalite kontrol çalışmalarının yapılması ile birlikte bitkisel ekstraktın in vitro olarak antioksidan özellik gösterip göstermediği incelenmiştir. Yöntem: Rheum ribes L. bitkisi 2016 yılında Metin Yıldırım tarafından Nisan-Mayıs aylarında toplanmıştır. Sert jelatin kapsüllere dolumu gerçekleştirilen Rheum ribes L. ekstraktlarında yığın dansitesi, sıkıştırılmış dansite, sıkıştırılabilirlik indeksi, Hausner oranı, akış süresi, yığın açısı tayini, elek analizi, dolumu yapılan sert jelatin kapsüllere ise dağılma testi, ağırlık sapması testi yapılmıştır. Bitki ekstraktının antioksidan aktivitesi 1,1-Difenil 2-Pikril Hidrazil (DPPH) metodu ile ölçülmüştür. Bulgular: Elde edilen sonuçlara göre yığın açısı 26.66±0.3o, yığın dansitesi 0.37±0.06 g/ml, sıkıştırılmış dansite 0.46±0.09 g/ml, elek analizinin sonucuna göre d50: 214.734±12.36 µm, d90: 43.99±2.92 µm bulunmuş olup, yapılan dağılma testinde ise suni mide vasatında 1 dakika 12 saniyede, suni barsak vasatında 2 dakika 10 saniyede kapsüller dağılma göstermiştir. Dolumları yapılan kapsüllerde ağırlık sapması 250.16±1.85 mg’dır. Rheum ribes L. ekstraktının IC50’si 74.1±4.8 µg /ml olarak bulunmuştur. Sonuç: Elde edilen verilere göre, Avrupa Farmakopesi baz alındığında kapsüllere dolum yapılan tozun akışkanlığı mükemmel, elek analizi sonucuna göre tozun inceliği çok kaba olarak bulunmuştur. Bu sonuçlara göre; Rheum ribes L. ekstraktı içeren sert jelatin kapsüllerin formülasyon açısından bir sorun teşkil etmediği ve piyasada gıda takviyesi olarak kullanılabileceği düşünülmektedir.

Quality Control of Hard Gelatin Capsules Containing Rheum Ribes L. Extract and Investigation of Its Effects on Oxidative Stress

Aim: Rheum species are important plants in terms of medicine. Rheum ribes L. is locally known as Işkın, Uçkun or Uşgun. It is usually grown in Lebanon, Iran and the east of Turkey. Extracts from various parts of the plant Rheum ribes L. are used in the treatment of many diseases. In this study, the preparation of Rheum ribes L. extract which is used for treatment of diabetes, hemorrhoids, ulcer and diarrhea, powder controls of the obtained extracts in accordance with the European Pharmacopoeia, obtaining hard gelatin capsule formulation of prepared extract and capsule quality control studies of hard gelatin capsules including Rheum ribes L. extract, it was investigated whether the herbal extract demonstrated antioxidant properties in vitro. Method: Rheum ribes L. were collected by Metin Yıldırım in April-May 2016. Hard gelatin capsules filled with Rheum ribes L. extracts were subjected to an angle of repose determination, bulk density, tapped density, sieve analysis, compressibility index, Hausner ratio, flow time and filled hard gelatin capsules were subjected to disintegration test and a weight deviation test. Antioxidant activity of plant extract was calculated by 1,1- diphenyl-2-picrylhydrazyl (DPPH) method. Results: According to the results obtained, the angle of repose was 26.66 ± 0.33 ̊, the bulk density was 0.37±0.06 g/ml, the tapped density was 0.46±0.09 g/ml, the particle size as d50: 214.734±12.36 µm, d90: 43.99±2.92 μm was exhibited by sieve analysis and the disintegration test presented disintegration in artificial gastric medium at the end of 1 minute 12 seconds, in artificial intestinal medium at the end of 2 minutes and 10 seconds and the weight deviation in filled capsules was 250.16±1.85 mg. The IC50 of Rheum ribes L. extract was found to be 74.1 ± 4.8 µg / ml. Conclusion: According to the data obtained, based on the European Pharmacopoeia, the flowability of the powder-filled into capsules was ‘excellent’ and the particle size of the powder was found to be ‘very coarse’ powder according to sieve analysis. Hereunder to these results; it is believed that hard gelatin capsules containing Rheum ribes L. extract are not a problem in the formulation and can be used as a food supplement in the market.
