In Vivo Pb(NO3)2 Toksisitenin Fizyolojik, Sitogenetik ve Anatomik Parametreler ile Araştırılması
Bu çalışmada, Pb(NO3)2’nin Allium cepa L.’da sebep olduğu fizyolojik,anatomik ve sitogenetik etkiler araştırılmıştır. Kök uzunluğu, çimlenmeyüzdesi ve ağırlık artışı fizyolojik parametreler; kromozomal hasaroluşumu, mitotik indeks (MI) ve mikronukleus (MN) sıklığı isesitogenetik parametreler olarak kullanılmış, kök ucu kesitlerinde iseanatomik hasarlar araştırılmıştır. Toksisitenin belirlenmesi amacıylabir kontrol ve üç uygulama grubu oluşturulmuş ve uygulama grupları50, 100 ve 200 mg L-1 Pb(NO3)2 çözeltisi ile 25 °C’de 72 saat muameleedilmiştir. Deneysel işlemler sonucunda, Pb(NO3)2 uygulamasınınçimlenme yüzdesi, kök uzunluğu, ağırlık kazanımını ve MI oranınıazalttığı, MN sıklığı ve kromozomal hasar oluşumunu arttırdığıbelirlenmiştir. Pb(NO3)2’nin fragment, yapışkan kromozom, kromozomköprüsü ve c-mitoz şeklinde kromozomal hasarları teşvik ettiğigözlenmiştir. Ayrıca, Pb(NO3)2 uygulamasının A. cepa kök ucundakorteks hücre çeperinde kalınlaşma, hücre deformasyonu, belirginolmayan iletim doku, nekroz gibi anatomik hasarlara neden olduğu datespit edilmiştir. Sonuç olarak, Pb(NO3)2’nin doza bağlı olaraktoksisiteye neden olduğu ve bu hasarların belirlenmesinde A. cepa’nıngüçlü bir biyoindikatör olduğu tespit edilmiştir.
Investigation of In-vivo Pb(NO3)2 Toxicity with Physiological, Biochemical and Cytogenetic Parameters
In this study, physiological, anatomical and cytogenetic effects of Pb(NO3)2 in Allium cepa L. were investigated. The root length, percentage of germination and weight increase were used as physiological parameters and chromosomal damage frequency, mitotic index (MI) and micronucleus (MN) frequency were determined as cytogenetic parameters and anatomical damages were investigated in root tip cross sections. To determine the toxicity, a control and three treatment groups were formed and the treatment groups were germinated with 50, 100 and 200 mg L-1 Pb(NO3)2 at 25°C for 72 hours. The results indicated that Pb(NO3)2 application decreased the germination percentage, root length, weight gain and MI, and increased the MN frequency and chromosomal damage formations. It was observed that Pb(NO3)2 induced chromosomal damages such as fragment, sticky chromosome, chromosome bridge and c-mitosis. In addition, Pb(NO3)2 application caused the anatomic damages including cortex cell wall thickening, cell deformation, nonspecific transmission tissue, necrosis in A. cepa root cells. As a result, Pb(NO3)2 was found to cause toxicity depending the application dose and it was determined that A. cepa was a strong bioindicator in determining the toxicity.
