L-arjinin ve N - nitro - L - arjinin metil esteri uygulamasının beyin, karaciğer, böbrek dokusu nitrik oksit ve malondialdehit düzeylerine etkisi

Bu çalışmada nitrik oksit (NO) vericilerinden L-arjinin ve NO sentezini inhibe eden N-nitro-L-arjinin metil ester (L-NAME)’inin uzun süreli uygulamasının beyin, karaciğer ve böbrek dokularındaki NO ve malondialdehit (MDA) düzeylerine etkisinin belirlenmesi amaçlandı. Araştırmada 3 farklı grup oluşturuldu ve kontrol grubuna 0.5 ml izotonik NaCl, L-arjinin grubuna 200 mg/kg L-arjinin ve L-NAME grubuna 100 mg/kg L-NAME günde bir kez olmak üzere 11 gün boyunca intraperitoneal yolla enjekte edildi. Denemenin sonunda ötenazi edilen tavşanların beyin, karaciğer ve böbrek dokuları alındı. Alınan doku örneklerinde NO ve MDA düzeyleri kolorimetrik yöntemle belirlendi. Karaciğer ve böbrek dokularındaki NO ve MDA düzeyleri açısından gruplar arasında istatistik olarak anlamlı bir fark bulunmazken, L-arjinin enjekte edilen grupta beyin dokusu NO ve MDA düzeylerinin diğer gruplara göre istatistik olarak yüksek (P

Effect of L - arginin and N - nitro - L - arginine metyl ester brain, liwer and kidney tissue nitric oxide and malondialdehyde levels

This study was aimed at evaluating brain, liver and kidney tissue NO and malondialdehyde (MDA) after long term administration of L-arginine a nitric oxide donor and N nitro-L-arginine methyl ester (L-NAME) a NO inhibitor. Three groups were established; control group received 0.5 ml isotonic NaCl, L-arginine group was given 200 mg/kg L-arginine and L-NAME group received 100 mg/kg L-NAME by intraperitoneal injection daily for 11 days. Rabbits were euthanized at the end of the experiment and brain, liver and kidney tissue samples were collected. Tissue NO and MDA levels were colorimetrically determined. There was no statistically significant difference in liver and kidney NO and MDA level between the groups while Larginine administered group had higher brain NO and MDA levels when compared to other groups (P<0.05). As a result, long term administration of L-arginine increased brain tissue NO and MDA when compared to liver and kidney values.
