Koyunlarda anaplasma phagocytophilum a karşı antikorların Western Blot yöntemi ile araştırılması

Koyunlarda kene hummasına, zorunlu hücre içi bir bakteri olan Anaplasma phagocytophilum neden olmaktadır. Bu çalışmada Kars bölgesinde 104 koyundan toplanan serum örnekleri organizmanin insan orijinli A, phagocytophilum izolatı kullanılarak Western blot yöntemi ile testi edildi. Değerlendirilen 12 örnekte (%11.5) A. phagocytophilum'un 61- ve 43-kDa antijenlerine, 8 örnekte (%7.7) sadece 43-kDa antijenine ve 7 örnekte ise (%6.7) sadece 61-kDa antijenine karşı reaktivite tespit edildi. Kars bölgesinde kene hummasının varlığını ortaya koyan bu çalışma, veteriner hekimlerin ve koyun yetiştiricilerinin bu hastalığı dikkate almaları gerekliliğini belirlemiştir. Ayrıca etkenin veya varyantlarının zoonotik potansiyeli nedeniyle hastalık beşeri hekimler tarafından da dikkate alınmalıdır.

Investigation of antibodies against anaplasma phagocytophilum in sheep by Western Blot analyses

Tick-borne fever in sheep is caused by an obligatory intracellular bacterium, Anaplasma phagocytophilum (formerly known as Ehrlichia [Cytoecetes] phagocytophila). One hundred and four sheep serum samples from different localities in Kars region were tested to detect antibodies against A. phagocytophilum by Western blotting using purified human-originated A. phagocytophilum. Twelve sera (11.5%) reacted with both 61- and 43-kDa antigens of A phagocytophilum while 8 sera (7.7%) reacted with 43-kDa antigen only. Seven (6.7%) serum samples had reactivities to 61-kDa of Anaplasma antigen. The present study indicates that A. phagocytophilum infection exists among sheep in Kars and this should be considered by both veterinarians and physicians in this region.
