Koyun Fötus derisinde Mast hücrelerinin gelişimi ve sayısal yoğunluğu

Bu çalışmada, fötal koyun derilerinde mast hücrelerinin histolojik ve histokimyasal özellikleri ve dağılımları incelendi. Fötuslar yedi yaş grubuna ayrıldı. Deri örnekleri 56. ve 145. günler arasında toplam 35 fötustan alındı. Doku örnekleri tamponlu formaldehitte tespit edildi. Hazırlanan parafin bloklardan 5 μm kalınlığında kesitler alındı. Örnekler hemotoksilen eosin, toluidin blue (TB) ve alcian blue-safranin O (AB/SO) ile boyandı. Işık mikroskopta incelendi. Mast hücrelerinin sayısal yoğunluğu birim alanda (1 mm2) belirlendi. Mast hücreleri ilk 83. günlük fötus derisinde (3. grupta) görüldü. Hücreler dermiste kan damarları ve kıl folikülleri çevresinde yerleşmişti. Mast hücrelerinin 3. ve 4. grupta AB pozitif, son üç grupta ise AB/SO pozitif granüller taşıdığı izlendi. Mast hücrelerinin yoğunluğu 3. grupta (75-85. günde) 2.80±0.87/mm2, dördüncü grupta (85-100. gün) 12.40±2.03/mm2, beşinci grupta (100-110. günde) 22.80±2.01/mm2, altıncı grupta (110-120. gün) 26.00±1.04/mm2 ve yedinci grupta (120-145. günde) 27.20±0.24/mm2 olarak belirlendi. Mast cell development and density in fetal sheep skin

In this study, histological and histochemical properties and distrubition of mast cells (MCs) were investigated in the fetal sheep skin. Fetuses were divided into 7 age groups. Skin samples were taken from 35 fetuses of between 56 day and 145 day-old. The samples were fixed in neutral buffered formaline and paraffin sections were cut a thickness of 5 &#956;m. Tissues were stained with haematoxylin and eosin, toluidine blue (TB) and alcian blue/safranin O (AB/SO). Mast cell numbers determined in a test area (1 mm2). Mast cells were firstly recognized in skin of 83 days-old fetuses. The MCs were observed around the blood vessels and hair follicles in dermis. MCs granules were stained with AB in 3rd and 4th groups also were stained with AB/SO in last three groups. MCs numbers were 2.80±0.87/mm2 in 3rd group (75th-85th days), 12.40±2.03/mm2 in 4th group (85th-100th days) 22.80±2.01/mm2 in 5th group (100th-110th days), 26.00±1.04/mm2 in 6th group (110th-120th days) and 27.20±0.24/mm2 in 7th group (120th-145th days). The significance between 3rd-4th groups and last three groups were statistically important (P<0.001).
