Topraktan İzole Edilen Streptomyces sp.’den Ksilanaz Üretimi ve Karakterizasyonu

Bu çalışmada topraktan izole edilen Streptomyces sp.’nin izolasyonu, tanımlanması ve optimizasyonugerçekleştirilmiştir. Streptomyces sp.’nin optimum şartları belirlenerek optimum 35 oC, pH 7.0 ve 52. saat olarakmaksimum üreme gerçekleştirdiği tespit edilmiştir. Ksilanaz üretiminin en iyi olduğu koşullar ise 35 oC, pH 7.0ve 72. saat olarak belirlenmiş ve en iyi enzim aktivitesine 40oC ve pH 7.0’de ulaşılmıştır. Enzim aktivitesi üzerinemetallerin etkisi araştırıldığında Zn 2+, Hg 2+ ve Fe 2+ elementlerinin güçlü bir şekilde enzim aktivitesini inhibeettiği gözlemlenmiştir. Kullanılan deterjanlardan Triton X-100 aktiviteyi arttırırken SDS ise güçlü şekilde inhibeettiği tespit edilmiş ve enzim aktivitesinin; 35oC-45oC aralıklarında da 1 saat boyunca stabil kaldığı, pH 6.0, ve 7.0de 1 saat sonunda %100 korunduğu görülmüştür

Production and Characterization of Xylanase from Streptomyces sp. which is Isolated from Soil

In this work, Streptomyces sp., was isolated from soil, identifed and optimized. Optimum conditionsof Streptomyces sp. were determined and it is observed that maximum reproduction is achieved at 35 oC, pH 7.0and 52 hours. It was also revealed that the best conditions for producing xylanase was 35 oC, pH 7.0 and 72 hoursand the best enzyme activity was achieved at 40 °C and pH 7.0. The effect of metals on enzyme activity wasinvestigated, and it was observed that enzyme activity was strongly inhibited by Zn 2+, Hg 2+ and Fe 2+. It was foundthat Triton X-100, one of the detergents was used, boosts activity, while other one, SDS, inhibits strongly and it isrevealed that, enzyme activity remains stable for 1 hour at 35oC, 40 oC and 45oC and also it is preserved 100% after1 hour at pH 6.0 and pH 7.0.
