Ovalbüminin titanyum dioksit ve zirkonya yüzeylerle etkileşimi ve adsorpsiyonunun araştırılması

Protein molekülleriyle biyomalzeme yüzeyleri arasındaki etkileşimlerin bilinmesi, biyomedikal teknolojiler vemalzeme mühendisliğinin birçok uygulamasında önem taşımaktadır. Protein makromoleküllerinin yapılarının veadsorpsiyonunun karmaşıklığı nedeniyle bu konudaki bilgi eksikliği, protein-yüzey etkileşimlerinin çözümlenmesine yönelik çalışmalara gereksinim duyulmasına yol açmaktadır. Proteinlerin adsorpsiyonuproteinlerin kendi özellikleri, pH, derişim, iyonik şiddet ve sıcaklık gibi birçok parametreden etkilenmektedir. Bu çalışmada Ovalbümin proteininin sulu çözeltilerden TiO2 veZrO2 üzerine adsorpsiyonunun incelenmesi,adsorpsiyona çözelti pH'ı, sıcaklığı ve adsorbent miktarının etkilerinin incelenmesi; adsorpsiyon hızı, proteinmetal oksit yüzey etkileşimlerinin ve adsorpsiyonu kontrol eden koşulların araştırılması amaçlandı. Ovalbüminproteininin sulu çözelti ortamında TiO2 ve ZrO2 tozları ile etkileşimleri ve adsorpsiyonu kesikli sistemde araştırıldı.Adsorpsiyon deneyleri pH 5, 6,4, 8 ortamında 20, 25, 30 °C'de yürütüldü. Ayrıca 0,5-1,0 g/L aralığında farklı miktarlarda TiO2/ZrO2 eklenerek, adsorbent miktarının etkisi incelendi. Bulgular, adsorplanan Ovalbüminmiktarının, sıcaklık ve adsorbent miktarının artmasıyla arttığını, pH ile azaldığını gösterdi. Adsorbent miktarının artmasıyla temas yüzeyinin artması, adsorplanan protein miktarının artmasına yol açmaktadır. Zeta potansiyelölçümleri, Ovalbümin ile TiO2 ve ZrO2 arasındaki etkileşimlerin, proteinin izoelektrik noktasına yakın olan pH5'de daha fazla olduğunu gösterdi. Ovalbüminin TiO2 ve ZrO2 üzerinde farklı sıcaklıklar ve çözelti pH'larındakiadsorpsiyonunun her iki adsorbent için de birinci mertebe kinetiği ile daha uyumlu olarak ilerlediği görüldü.

Investigation of the interactions and adsorption of ovalbumin with titanium dioxide and zirconia surfaces

Knowing the interactions between the protein molecules and the biomaterial surfaces is important in a number ofapplications in biomedical technologies and materials engineering. The lack of knowledge on this topic due to thecomplexity of the protein molecular structures and their adsorption necessitates carrying out research work in orderto solve the problems related with the protein-surface interactions. Protein adsorption is influenced by manyparameters such as the properties of proteins, pH, concentration, ionic strength and temperature. The aim of this study is to investigate the Ovalbumin adsorption from aqueous solutions on TiO2 andZrO2, to study the effects of solution pH, temperature and adsorbent dosage on adsorption; to investigate the adsorption kinetics, protein-metal oxide interactions, and to determine the conditions controlling the adsorption. The interactions and adsorption ofOvalbumin protein with TiO2 and ZrO2 powders in solution environment were investigated in a batch adsorptionsystem. Adsorption experiments were carried out in pH 5, 6.4, 8 environment at 20, 25, 30 °C. The effect ofadsorbent dosage was also investigated by the addition of different amounts of TiO2/ZrO2 in the range 0.5-1.0 g/L. Results have shown that the adsorbed amount of Ovalbumin increases with increasing temperature and adsorbent dosage, but decreases with increasing pH. The increase of contact surface with increasing adsorbent dosageincreases protein adsorption. The zeta potential measurement results supported that the interactions betweenOvalbumin with TiO2 and ZrO2 were higher at pH 5, which is close to the isoelectric point of protein. Theadsorption of Ovalbumin onto TiO2 and ZrO2 at different temperatures and solution pH conditions followed firstorder kinetics with a better fit for both adsorbents.
