Bu çalışmada miniprep ve ultrasantrifüj yöntemiyle plazmid izolasyonları incelenmiştir. Standart bakteri ve onun bilinen plazmidleriyle yapılan plazmid izolasyonunda miniprep yönteminin çeşitli problemleri olduğu görülmüştür. Özellikle miniprep yönteminde görülen 80 kb civarındaki fazla bant ultrasantrifüj yönteminde görülmemektedir. Miniprep yöntemi içerdiği bu fazla bant veya bantlar ile daha ileri moleküler çalışmalar için sakınca teşkil etmektedir.

In this study isolated plasmids were examined using miniprep and ultracentrifugal methods. It was observed that there were some kinds of problems related to miniprep procedure for plasmid isolation from standard bacteria and its known plasmids. Particularly, an extra band close to 80 kb size seen in miniprep procedure did not appear in ultracentrifugal method. Miniprep procedure has a limitation for further molecular investigations because of this/these extra band(s).
