Yaşlanan olfaktor mukozadaki değişimlerin ince yapı düzeyinde araştırılması
Amaç: Bu çalışmada, doğumdan sonra geç erişkin ve yaşlılık dönemine kadar farklı yaş gruplarında, olfaktör mukozada olaylanan yapısal değişimlerin elektron mikroskobik düzeyde araştırılması amaçlanmıştır. Gereç ve Yöntem: Bu amaçla, yeni doğan, 5 haftalık, 7 haftalık, 3 aylık, ve 1 yaşındaki dişi 15 adet Wistar-albino sıçan her grupta 3'er denek olacak şekilde gruplandırıldı. Alışılagelmiş elektron mikroskobik takip yöntemlerinden geçirilen dokulardan alman yarı ince kesitler toluidine mavisi ile boyandıktan sonra fotoışık mikroskobunda değerlendirildi. İnce kesitler ise Cari Zeiss EM 900 ile değerlendirilip fotoğrafları çekildi. Bulgular: Olfaktör mukoza epitelyuminin, tüm gruplarda üç tip hücre kapsadığı belirlendi. Lamina propriya ise kan damarları, bağ dokusu lifleri, Bowman bezleri içeren gevşek bağ dokusu yapısmdaydı. Yeni doğan ve gençlik döneminde olfaktör epitelde yapısal olarak bir değişim izlenmezken yaşlanma ile birlikte özellikle olfaktör reseptör hücrelerinin silyalarmda olmak üzere, destek hücrelerinin mikrovilluslarmda ve tüm mukozada belirgin yapısal dejenerasyon belirlendi. Ayrıca apoptotik hücrelerde de yaşla birlikte artış izlendi. Sonuç: Sonuç olarak yaşlılık organ, doku, hücre ve organellerin aktivitelerini yavaşlatan karmaşık bir süreçtir. Bu nedenle olfaktör mukozada da yaşın ilerlemesine koşut dejeneratif değişimlerin ortaya çıkması doğaldır kanısına varılmıştır.
Ultrastuctural investigation of aging olfactory mucosa
Purpose: To investigate ultrastractural changes in different age groups, from birth to adult ages and in the senescence period. Methods: Fifteen Wistar albino rats were used, in five groups (group 1: newborn; group 2: 5 weeks old; group 3: 7 weeks old; group 4: 3 months old; group 5: 1 year old), with five animals in each. Semi-thin sections were stained with Toluidine blue and evaluated under a photo-light microscope after routine electron microscopic processing had been done. Thin sections were investigated and estimated by Carl Zeiss EM 900. Results: Three types of cell were determined in the olfactory epithelium in all groups. Lamina propria was a loose connective tissue that included blood vessels, connective tissue fibers, and Bowman's glands. We did not observe any degenerative changes in the newborn or young groups, although considerable degeneration in the cilia and microvilli of olfactory neurons and sustentacular cells was seen in the elderly group's olfactory mucosa. Apoptotic figures and cells were seen in the elderly group also. Conclusion: Senescence is a complicated period that decelerates the activities of organelles, tissues, and cells. For this reason, the degeneration and changes, determined in our study in olfactory mucosa were considered normal.
