Kiçük hücreli dışı akciğer karsinomlarında nükleer faktör kappa B İmmünohistokimyasal ekspresyonunun prognozla ilişkisi

Amaç: Son dönemde tanımlanan bir transkripsiyon faktörü olan Nükleer Faktör kappa B (NF-kB) ile ilgili literatürde çok sayıda çalışma bulunmaktadır. Bu çalışmalarda çeşitli tümörlerde immünhistokimya ile nükleer ve/veya sitoplazmik boyanmanın olduğu ve hasta prognozu ile ilişki gösterdiği ileri sürülmektedir. Çalısmamızda NF-kB ekspresyonunun küçük hücreli dışı akciğer karsinomlarırida prognoz ile ilişkisini araştırmayı amaçladık. Gereç ve Yöntem: Çalışmamızda bu protein ailesinin iki önemli üyesi olan p50 ve p65'in (RelA) 30'u klinik izlemli 52 küçük hücreli dışı akciğer karsinomu hastasmdaki immünhistokimyasal ekspresyonunu ve bunun prognozla ilişkisini inceledik. Bulgular: Çeşitli histopatolojik tanı gruplarındaki olgular immünhistokimyasal olarak p50 ve p65 ile yüksek oranda sitoplazmik ekspresyon göstermiştir. Nükleer boyanma düşük oranda saptanmıştır. Her iki antikor ile klinik parametreler ve sağkalım arasında istatistiksel olarak anlamlı sonuç elde edilememiştir. p50 ve p65'in hem nükleer hem sitoplazmik boyanma yoğunluk ve yaygınlıkları arasında ise anlamlı pozitif korelasyon saptanmıştır. Sonuç: Küçük hücreli dışı akciğer karsinomları immünhistokimyasal olarak NF-kB p50 ve p65 komponentlerini yüksek oranda eksprese etmektedir. Ancak bu ekspresyon prognostik faktörlerle ilişkili bulunmamıştır.

Prognostic significance of nuclear factor kappa B immunohistochemical expression in non-small cell lung carcinomas

Purpose: To investigate the immunohistochemical expression and the prognostic significance in non-small cell lung carcinomas of a recently defined. transcription factor, Nuclear Factor kappa B (NF-kB), which has been reported to be expressed in the nucleus and/or cytoplasm by a variety of neoplasms in the literature. Materials and Methods: Immunohistochemistry against the two major components of this protein family, p50 and p65 (RelA), was performed on 52 patients' material diagnosed as non-small cell lung carcinoma, for 30 of which we had the clinical follow-up data. Results: We observed a high rate of cytoplasmic p50 and p65 expression in all different lung carcinoma histological subtypes. Nuclear staining was only found in a few cases. There was no statistical significance'between staining rates and clinical parameters or patient survival. A positive correlation was shown between nuclear and cytoplasmic staining rates of both p50 and p65. Conclusion: Non-small cell lung carcinomas highly express NF-kB p50 and p65 components immunohistochemically. Nevertheless, this expression does not significantly affect prognosis.
