İnsan ve keçi serumundaki lipoproteinlerin fosfotungstik asit $ASİT/MgCI_2$ presipitasyon metodu ile separasyonunun karşılaştırılması

Comparison of separation of lipoproteins in human and goat serum by phosphotungstic Acit/MgCI_2$ precipitation method
Lipoproteins are water-soluble complexes through which fats and lipids are transported between the various tissues for storage and utilization. Association of lipoproteins with risk of coronary heart disease and various types of dyslipoproteinemias 'necessitates the quantification of the individual lipoprotein classes. One of the most commonly used techniques for the separation of $beta$ and pre- $beta$ lipoproteins is the precipitation by phosphotungstic acid / $MgCl_2$ - However, none of %e lipoprotein separation techniques used in laboratories has been developed for use in animal lipoproteins. We, therefore, undertook the present study to compare the phosphotungstic acid / $MgCl_2$ concentrations required for the precipitation of apolipoprotein-B containing lipoproteins ( $beta$ and pre- $beta$ ) in both human and goat senimin accordance .with the orginal method, we confirmed by cellulose-acetate electrophoresis that 0.56 |imol / 25 umol phosphotungstic acid / $MgCl_2$was sufficient for the precipitation of apolipoprotein-B containing lipoproteins in human serum. Whereas goat serum required approx. 3 times more phosphotungstic acid / $MgCl_2$(min. 1.67 $mu$ mol/75 $mu$ mol; max. 1.97 $mu$ mol/87.5 $mu$ mol) for the precipitation of the same lipoprotein classes than did human serum. Addition of 4.44 $mu$ mol / 200 $mu$ mol phosphotungstic acid / $MgCl_2$ resulted in the complete precipitation of all the lipoprotein classes of goat serum. Possible causes of the difference in the concentration of phosphotungstic acid / $MgCl_2$required for the precipitation of apolipoprotein-B containing lipoproteins in human and goat serum were discussed.
