Bands were observed in ultraviolet lamps for CA 1 and appropriate light condition for ME.Results: No polymorphism was observed for the CA 1 enzyme. All sheep were found to be homozygote for the same allele. For ME loci, population were observed polymorphism, having F, FS and S bands, the difference between F and S bands in ME loci were unimportant (p>0.05) and allels were determined in the Hardy-Weinberg equilibrium.Conclusion: In this study, ME polymorphism was observed but no polymorphism was determined for CA 1 locus. It was concluded that it is necessary to investigate other enzymes in terms of polymorphism. Acıpayam koyunlarında karbonik anhidraz 1 (CA1) ve malik enzim (ME)'in nişasta jel elektroforezi ile araştırılması

Amaç: Acıpayam ırkı koyunların genetik yapılarını belirlemek için iki enzim lokusu; karbonik anhidraz (CA 1] ve malik enzim (ME] nişasta jel elektroforezi ile incelemektir.Gereç ve Yöntem: Araştırmada "Denizli Acıpayam Devlet Üretme Çiftliği"nden 50 adet koyundan kan alındı. Elde edilen hemolizatlar elektroforez işlemleri uygulanarak incelendi. Her iki enzim için Tris-EDTA-Borat pH:8.6 tamponu kullanıldı. Elektroforez sonrası boyamada CA 1 için MUA, aseton, agar noble, fosfat tamponu pH:6.5 ve ME için NADP, MTT, PMS, DL Malik asit, MgC12, NaOH, Agar Noble, Tris-HCl pH:8.0 tamponu kullanıldı. Bantlar CA 1 için ultraviyole ışıkta, ME için ise uygun ışık ortamında gözlendi.Bulgular: CA 1 enzimi lokusları yönünden koyunlarda po-limorfizm tespit edilmedi ve bütün numunelerin aynı bantlara sahip homozigot bireyler olduğu görüldü. ME lokusu yönünden populasyon içi polimorfizm gözlendiği, F, FS ve S bantlarına sahip olduğu, ME lokusunun F ve S allel frekans değerleri arasındaki farkın önemsiz olduğu (p>0.05) ve al-lellerin Hardy-Weinberg dengesinde olduğu belirlendi..Öneri: Bu çalışmayla Acıpayam koyunlarında malik enzim yönünden polimorfizm görülürken, CA 1 yönünden polimorfizm görülmediği belirlenmiş ancak başka enzimler açısından da polimorfizm gösterip göstermediğinin araştırılması gerektiği sonucuna varılmıştır.
