Türkiye'de yetiştirilen holştayn ve bazı yerli sığır ırklarında citrullinemia allelinin belirlenmesi
Bu çalısmanın amacı Türkiye’de yetistirilen Holstaynlarda ve Türkiye yerli sıgır ırklarında citrullinemia allelinin bulunup bulunmadıgının arastırılmasıdır. Çalısmanın materyalini, 176 bas Holstayn sıgırı ile 10 bas Yerli Kara, 10 bas Boz Irk, 10 bas Güney Anadolu Kırmızısı ve 10 bas Dogu Anadolu Kırmızısı sıgırı olusturmustur. Hedef bölge PCR ile çogaltılmıs ve pozitif örnekler AvaII restriksiyon enzimiyle sindirime tabi tutulmustur. AvaII polimorfizminin varlıgı % 3’lük NuSieve agaroz jel elektroforezi ile kontrol edilmistir. Incelenen Holstaynlarda ve yerli sıgırlarda citrullinemia tasıyıcısına rastlanılmamıstır.
The detection of citrullinemia allele in holstein and some native Turkish cattle breeds
The aim of this study was to investigate whether the citrullinemia allele exists in Holsteins and in Turkish native cattle breeds in Turkey. The material of the study consisted of 176 Holstein cattle and 10 Native Black, 10 Gray Steppe, 10 South Anatolian Red and 10 East Anatolian Red breeds of cattle. The target site was amplified by PCR and positive samples were digested by AvaII restriction endonuclease enzyme. AvaII polymorphisms were controlled in 3% NuSieve agarose gel electrophoresis. There were no positive results indicating the presence of citrullinemia allele in Holsteins and Turkish native cattle breeds.
