Diabetik hasta serumunun sıçanda Schwann hücre canlılığı üzerine in vitro etkisi

Amaç: Schwann hücresi (SH) periferik sinirlerde aksonların miyelin kilifinın yapiminı saglayarak sinir fonksiyonunu destekleyen glial hücredir. Diabetik nöropatide SH kaybı ya da hasarı gözlenmektedir. Daha önceki çalismalar diabetik hasta serumunun arka kok ganglion noron kültüründe hücre ölümüne yol actigiru göstermiştir. Diabetik hasta serumunun SH üzerine benzer bir etkisinin olup olmadigini arastirmak amaciyla bu çalışmada diabetik hasta serumunun in vitro sican SH cankkgi uzerine etkisi degerlendiriknistir. Gereç ve yöntem: Bu calışmada sican siyatik sinirinden hazirlanan SH kültürlerine diabetik hasta serumu ve normal kontrol serumu lsiyla inaktivasyon sonrasi degisik oranlarda eklendi. Hücre canlılıginin değerlendirilmesinde 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) testi kullanildi. Bulgular: Normoglisemik kontrol kiiltiirleriyle karsilastirildiginda diabetik hasta serumu eklenen SH kültürlerinde hücre canlılıginin istatistiksel olarak anlamli fark gostermedigi saptandi. Sonuc;: Bu sonuclar lsiyla inaktive edilmis diabetik hasta serumunun sican SH kültürleri üzerine anlamli bir toksisitesinin olmadigini gostermektedir.

The effect of diabetic patient serum on rat Schwann cell viability in vitro

Schwann cells (SCs) are gkal cells which are responsible for the myelin production and supporting nerve function in peripheral nerve system. SC loss or in¬jury occur in diabetic neuropathy. Recent studies have reported that diabetic patients serum leads cell death in dorsal root ganglion neuron culture. To evaluate whether diabetic patient serum has similar effect on SC viability, the effect of patient serum on SC viabikty has been determined in SC cultures, Material and method: In the present study, diabetic patient serum and normal control serum were added to SC cultures that were prepared from rat sciatic nerve. 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) test was used to evaluate cell viability. Results: It was found that diabetic patient serum did not induce significantly cell death in SC cultures as compared with control cultures. Conclusion: The results of the present study show that diabetic patient serum does not result significant cytotoxicity in rat SC cultures.
