Farklı fiziksel koşullarda Entamoeba histolytica’nın in vitro kültüründe elde edilen bulgular

Amaç. Bu çalışmada, deneysel ve antijen sağlamak amaçlı olarak yapılan Robinson ve Locke-Egg-Serum (LES) besiyerlerindeki kültürlerinde, sıcaklık, CO2 ve pH değişikliklerinden oluşan farklı fiziksel koşulların Entamoeba histolytica trofozoitleri üzerine etkilerinin belirlenmesi amaçlanmıştır. Yöntem. Subkültürleri Robinson ve LES besiyerinde 35,5 ºC’ de standart etüvde inkübe edilerek sürdürülen E. histolytica Bangladeş suşu kültürlerine paralel olarak 37,5 ºC’ de ve %5 CO2’li etüvde inkübasyon koşulları araştırılmıştır. Ayrıca pH 6,5 ve 7,5 olan besiyerlerinde yapılan kültürlerde in vitro çoğalma özellikleri izlenmiştir. Bulgular. Trofozoitlerin 37,5 ºC’ de canlılığını koruduğu ancak daha yavaş çoğaldığı gözlenmiştir. %5 CO2’li etüv ile standart etüv arasında ise belirgin bir fark saptanamamıştır. pH değişiklerinde ise 4. günde besiyerinde canlı parazit kalmadığı belirlenmiştir. Sonuçlar. E. histolytica’nın üretilme koşullarında farklılıklar denendiğinde en fazla pH değişikliğinden etkilendikleri, %5 CO2’li etüvün E. histolytica inkübasyonunda standart etüvün yerine kullanılabileceği görülmüştür.

The findings from in vitro Entamoeba histolytica culture under different physical conditions

Aim. Cultivation of Entamoeba histolytica is usually performed for diagnostic and experimental purposes. In this study, it is aimed to determine the effects of different physical conditions on the growth of E. histolytica Bangladesh strain in Robinson and Locke Egg Serum (LES) media. Methods. Parallel to regular passages at 35.5ºC in Robinson and LES media, incubation conditions for the 37.5ºC and %5 CO2 incubator is investigated. Additionally, the growth rates were further investigated in vitro at 6.5 and 7.5 pH values. Results. It was observed that at 37.5ºC trophozoites were able to survive but reproduction rate was decreased. Five percent CO2 incubator and standard bacteriological incubator did not yield difference in terms of parasite reproduction. No living trophozoite was detected in pH changed media on fourth day after passage. Conclusions. Among the changes in the cultivation of E. histolytica, pH has a crucial effect on parasite growth and %5 CO2 incubator can be used instead of standard incubator for incubation.
