17-β estradiole ile indüklenen insan lenfositlerindeki mikronükleus sıklığının incelenmesi

Bazı epidemiyolojik ve deneysel çalışmalar, kanser gelişimi ile östrojenler arasında bir ilişkinin olduğunu göstermiştir. Estrojenler ile ilişkili karsinogenezin, reaktif metabolitlerle direk DNA hasarı ve reaktif oksijen metabolitleriyle indirek genotoksik etkinin bir kombinasyonunun sonucu olduğunu düşündürmektedir. İnsan periferal kan lenfositlerinde uygulanan 'Sitokinezblok' mikronükleus (MN) metodu, genetik toksikoloji testleri arasında standart bir sitogenetik test olarak dünya üzerindeki birçok laboratuarca kabul edilen bir metodtur. Sunulan çalışmanın amacı, lenfosit kültürlerinde estradiol (E2)'ün muhtemel genotoksik etkilerini 'Sitokinez-blok' MN metodu kullanarak araştırmaktır. Çalışma, 50 sağlıklı erkek donörden izole edilen lenfositlerde gerçekleştirildi. Kontrol kültürleriyle (1-15 MN / 1000 hücre) karşılaştırıldığın da E2 (36 μM) uygulanan kültürlerde(1-11 MN /1000 hücre) MN sıklığında herhangi bir istatistiksel fark tespit edilmedi (p > 0.05). Ayrıca binükleuslu hücredeki MN sıklığı, E2 uygulaması ile değişmedi (Kontrol kültürlerinde: 5.88 ± 2.9; E2 kültürlerinde 5.64 ± 2.5, p > 0.05). Sonuçlarımız; estradiolün kültüre edilmiş lenfositlerde herhangi bir genotoksik etkisinin olmadığını göstermiştir.

17- β estradiole induced micronucleus formation in human lymphocytes

Some epidemiological and experimental studies showed that there is a relation between oestrogen and development of some cancers. The carcinogenicity of estrogens is considered to be the result of a combination of DNA damage by reactive metabolites and indirect genotoxicity by redox cycling and production of reactive oxygen species. Cytokinesis-block mikronucleus test (CBMN) in human peripheral blood lymphocytes has been accepted by many laborories all over the world as a standart cytogenetic investigation for testing the genetic toxicology. The objective of the present study was to evaluate possible genotoxic effects of estradiol (E2) in cultured lymphocytes, using the CBMN assay. This study was performed in human peripheral blood lymphocytes from a group of 50 healthy men. No statistically significant difference in micronucleus frequency was detected in E2 (36 μM) treatment (1-11 MN / 1000 cells) cultures compared to control(1-15 MN / 1000 cells) cultures (p > 0.05). The frequency of MN in binucleate lymphocytes was not altered by E2 treatment(5.88 ± 2.9 in control cultures and 5.64 ± 2.5 in treatment cultures; p > 0.005). Our results suggest that there are not any genotoxic effects of estradiol in cultured lymphocytes.
