Kolon Kanseri Hücresi Kültürü Ortam Değişikliği ve Proliferasyonuna Etkisi

Amaç: Çalışmada, laboratuvarda rutin olarak kullanılan ve yeni temin edilen hücre kültür vasatlarının ve serumlarının karşılaştırılması için hazırlanan kültür ortamlarının, HT-29 insan kolon kanser hücreleri üzerine etkilerinin incelenmesi amaçlanmıştır. Gereç ve Yöntem: Dört farklı kültür vasatı ile oluşturulan ortamda HT-29 insan kolon kanser hücreleri kültüre edildi ve proliferasyon belirteci Ki- 67 ve apoptoz belirteci Kaspaz-3 dağılımları immunohistokimyasal olarak incelenir iken, in vitro yara modelinde farklı ortamın etkileri morfolojik olarak değerlendirildi. Bulgular: Grup 1, Grup 2 ve Grup 3 HT-29 hücrelerinde Ki-67 immunoreaktivitesinin kuvvetli (+++) pozitif, Grup 4 HT-29 hücrelerinde ise orta şiddette (++) pozitif olduğu izlendi. Tüm grup HT-29 hücrelerinde aktif Kaspaz-3 immunoreaktivitesinin zayıf (+) şiddette olduğu tespit edildi. In vitro yara modelinde farklı kültür ortamlarının yara iyileşmesi üzerine etkisinin benzer olduğu görüldü. Sonuç: Çalışmada kullanılan farklı vasatların HT-29 insan kolon kanser hücrelerinin proliferasyonunu ve hücre ölümünü etkilemediği, yara modeli gruplarında ise benzer sonuçlar bulunması üzerine, özellikle farklı kültür ortamından ziyade, aynı marka ve serideki ürünlerin aynı anda kullanılmasının önemli olduğu sonucuna varıldı.

Objective: In this study, it was aimed to investigate the effects of culture media prepared for comparison between routinely used in the laboratory and newly obtained cell culture media and serum on HT-29 human colon cancer cells. Methods: HT-29 human colon cancer cells were cultured in four different culture media, and the proliferation marker Ki-67 and the apoptotic marker Caspase-3 distributions were examined immunohistochemically, while the effects of different media in the in vitro wound healing model were evaluated morphologically. Results: Ki-67 immunoreactivities were observed that in Group 1, Group 2 and Group 3 HT-29 cells were strongly (+++) positive and in Group 4 HT-29 cells were moderate (++) positive. It was determined that active Kaspase-3 immunoreactivity was weak (+) in all group HT-29 cells. It has been observed that the effect of different culture media on wound healing was similar in the in vitro wound healing model. Conclusion: It was concluded that the different media used in the study did not affect the proliferation and cell death of HT-29 human colon cancer cells, and that similar results were obtained in groups in the wound healing model. Also we demonstrate that using the same brand and same series products at the same time is important for utilization different culture media.
