Bulutlanma Noktası Ekstraksiyonu ile Zenginleştirildikten Sonra Ponceau 4R’nin Şurup ve Meyve Şekerinde Spektrofotometrik Tayini

Ponceau 4R (E-124) özellikle gıda ve ilaç sanayisinde sıklıkla kullanılan bir gıda boyasıdır. Bu çalışmada,Ponceau 4R’nin bulut noktası ekstraksiyonu için non iyonik bir yüzey aktif madde olan Brij 58 kullanılarak önderiştirme ve ayırma işlemi gerçekleştirilmiştir. Bu ön deriştirme işlemi sonrasında Ponceau 4R spektrofotometriktayini yapılmıştır. Bulutlanma noktası ekstraksiyonu sonrasında ayrılan yüzey aktif maddece zengin fazdakiPonceau 4R, su ile 0.5 mL'ye seyreltilmiş ve köre karşı 510 nm'de UV-görünür spektrofotometri ile absorbansıölçülmüştür. Ekstraksiyon için optimumum şartlar pH 6-9 aralığında, Brij 58 miktarı % 0.5 (w/v), tuz miktarı %20 (w/v), inkubasyon sıcaklığı 70 °C ve 10 dk bekleme süresi olarak belirlenmiştir. Yöntemin tayin aralığı 0.08 -2.50 g mL-1 olarak belirlenmiştir. Bu yöntemin saptama sınırı (LOD), 0.00453 g mL-1 olarak bulunmuştur. Bugeliştirilen seçici ve hassas bulutlanma noktası ekstraksiyonu yöntemi, Ponceau 4R’nin rutin analizi içinkullanılabilir.

Spectrophotometric Determination of Ponceau 4R in Syrup and Fruit Candy after Enrichment with Cloud Point Extraction

Ponceau 4R (E-124) is a food dye that is commonly used in the food and pharmaceutical industry. In this study, pre-concentration and separation was performed using Brij 58, a non-ionic surfactant, for cloud point extraction of Ponceau 4R. After this pre-concentration procedure, Ponceau 4R spectrophotometric determination was performed. After the cloud point extraction, the surfactant-rich phase Ponceau 4R was diluted to 0.5 mL with water and the absorbance was measured by UV-visible spectrophotometry at 510 nm against to blank. The optimum conditions for extraction were determined as in the pH range of 6-9, the amount of Brij 58 was 0.5% (w/v), the salt amount was 20% (w/v), the incubation temperature was 70 ° C and the 10 min waiting time. The determination range of the method was 0.08 - 2.50 g mL-1 . The detection limit (LOD) of this method was found to be 0.00453 g mL-1 . This developed selective and sensitive cloud point extraction method can be used for routine analysis of Ponceau 4 R.
