NANO GÜMÜŞ KATKILI Moringa oleifera L. (Brassicales: Moringaceae) SU EKSTRAKTININ Meloidogyne incognita (Kofoid & White, 1919) Chitwood, 1949 (Nematoda: Meloidogynidae) KARŞI LABORATUVAR KOŞULLARINDA ETKİNLİĞİNİN BELİRLENMESİ
Kök–ur nematodları (KUN) (Meloidogyne spp.), dünya genelinde tarımsal üretim yapılan yerlerde yaygınolarak bulunan ve geniş bir konukçu yelpazesine sahip önemli verim kayıplarına neden olanzararlılardandır. Dünyada Meloidogyne arenaria, M. exigua, M. graminicola, M. hapla, M. incognita, M.javanica ve M. mayaguensis önemli türler arasında yer almaktadır. Ülkemizde M. incognita, M. arenaria,M. javanica ve M. hapla türleri en yaygın bulunan türler olup M. incognita ve M. javanica ekonomikanlamda örtüaltı bitki yetiştiriciliğinde ciddi sorunlara neden olan türlerin başında gelmektedir.Nematisitler KUN’leri uzun süreli baskılayamazlar, çevresel ve insan sağlığı kaygıları kullanımlarıüzerinde artan kısıtlamalar ile sonuçlanmaktadır. Bu çalışma da, M. incognita’nın ikinci dönem larvaları(L2)’na karşı nano gümüş katkılı (AgNPs) Moringa oleifera L. (Brassicales: Moringaceae)’nın su ekstraktınematisidal etkinlik bakımından değerlendirilmiştir. Nano gümüş katkılı M. oleifera su ekstraktı, 42 ppm(0.25 mM), 84 ppm (0.50 mM) ve 168 ppm (1 mM) konsantrasyonlarda ve 5 tekerrürlü olarak in vitrokoşullarda denenmiştir. Denemede kullanılan her bir doz için petri kabına 1 ml L2 + 3 ml saf su +1 mlekstrakt eklenmiştir. Negatif kontrol olarak ise ekstrakt yerine saf su kullanılmıştır. Tüm petri kapları28±2℃’de muhafaza edilmiştir. Denemeden elde edilen sonuçlara göre bütün konsantrasyonlarda farklıoranlarda da olsa nematisidal aktivite gözlenmiştir. Nano gümüş katkılı M. oleifera su ekstraktının 168 ppm(1 mM) konsantrasyonu 72 saat sonunda %92.40 ölümüne sebep olduğu belirlenmiştir. Nano gümüş katkılıM. oleifera su ekstraktının tarımsal alanlardaki zararlı organizmalara karşı daha fazla sayıda denemeyapılarak elde edilen olumlu sonuçlar ışığı altında saksı ve sera koşullarındaki etkinliklerinin araştırılmasıönem arz etmektedir.
DETERMINATION OF THE EFFECTIVENESS OF NANO SILVER ADDITIVE AQUEOUS EXTRACT OF Moringa oleifera L. (Brassicales: Moringaceae) AGAINST ROOT–KNOT NEMATODE (Meloidogyne incognita (Kofoid & White, 1919) Chitwood, 1949 (Nematoda: Meloidogynidae)) UNDER LABORATORY CONDITIONS
Root–knot nematodes (RKNs) (Meloidogyne spp.) are widely distributed and cause significant yield losses in a wide range of crops. Meloidogyne arenaria, M. exigua, M. graminicola, M. hapla, M. incognita, M. javanica and M. mayaguensis are important species in the world. In Turkey, the species M. incognita, M. arenaria, M. javanica and M. hapla are the most commonly found, with M. incognita and M. javanica which causes serious problems to several economically important greenhouse crops. Nematicides do not provide long–term suppression of RKNs, and environmental and human health concerns are resulting in increased restrictions on their use. In this study, experiments were conducted to evaluate the effect of silver nanoparticles (AgNPs) using aqueous extract of Moringa oleifera L. (Brassicales: Moringaceae) at concentrations of 42 ppm (0.25 mM), 84 ppm (0.5 mM) and 168 ppm (1 mM) as a potential nematicide on second stage juveniles (J2s) of M. incognita were evaluated in vitro tests. The study was established according to randomized parcel design with 5 replicates. 1 ml of J2s suspensions + 3 ml of distilled water + 1 ml of extract were transferred into sterilized petri dishes. Distilled water was used as a negative control. All dishes were kept at 28±2℃. All concentrations showed different level of nematicidal activity. Silver nanoparticles using aqueous extract of M. oleifera at concentrations of 168 ppm (1 mM) was caused 92.40% nematodes mortality in 72 hours. In light of the positive results obtained by experimenting with more than the harmful effects of silver nanoparticles (AgNPs) using aqueous extract of M. oleifera in the agricultural areas, it is important to test the effectiveness in pots and greenhouse conditions.
