Kendinden bağlanabilen yeni bir akışkan kompozitin sitotoksisitesinin dentin bariyer testi ile değerlendirilmesi

AMAÇ: Bu çalışmanın amacı kendinden bağlanabilen yeni bir akışkan kompozitin (Vertise Flow) sitotoksisitesini, sığır dental pulpasından elde edilmiş hücrelerin üç-boyutlu kültürlerinin kullanıldığı bir dentin bariyer test düzeneği ile değerlendirmektir. GEREÇ VE YÖNTEM: Yapay hücre kültürü perfüzyon odaları 500 µm kalınlığındaki dentin diskleri ile iki kompartımana ayrıldı. Alt kompartımanda, üç-boyutlu hücre kültürleri dentine temas edecek şekilde yerleştirilerek pulpa odası kısmı oluşturuldu. Üst kompartımanda ise test materyalleri, dentin diskinin kavite kısmı üzerine üretici firmanın önerdiği şekilde uygulandı. Negatif ve pozitif kontrol materyalleri sırası ile President ve Vitrebond idi. Daha sonra, pulpal kısımdaki hücre kültürlerinin kültür ortamı ile perfüzyonları sağlandı. Yirmi dört saat sonra hücre canlılığı MTT testi ile değerlendirildi. Veriler Kruskal Wallis ve Mann Whitney U testleri kullanılarak istatistiksel olarak analiz edildi. BULGULAR: Vertise Flow ve Vitrebond (pozitif kontrol) gruplarının hücre canlılık oranları arasındaki fark istatistiksel olarak anlamlı idi (p < 0.05). Ancak, Vertise Flow ile President (negatif kontrol) gruplarının hücre canlılık oranları arasındaki fark istatistiksel olarak anlamlı değildi (p > 0.05) SONUÇ: Vertise Flow, sığır dental pulpasından elde edilmiş hücrelerin canlılığı üzerine negatif kontrol grubuna benzer bir etki gösterdi. Kalan dentin tabakası kalınlığının en az 0.5 mm olması durumunda, Vertise Flow’dan salınması muhtemel biyololojik olarak aktif bileşenlerin pulpa hücrelerini etkilemeyebileceği düşünülmektedir.

Cytotoxicity evaluation of a new self-adhering flowable composite by dentin barrier test

OBJECTIVE: The aim of this study was to evaluate the cytotoxicity of a new self-adhering flowable composite (Vertise Flow) in a dentin barrier test device using three-dimensional cultures of bovine dental pulp derived cells.MATERIALS AND METHOD: Artificial cell culture perfusion chambers were separated into two compartments using a 500 µm dentin disc. In the lower compartment, a pulp chamber was created by placing the three-dimensional cultures onto the dentin disc. Test materials were introduced into the upper compartment in direct contact with the cavity side of the dentin discs following the manufacturers’ directions. Negative and positive controls were President and Vitrebond, respectively. Subsequently, the pulpal part of the perfusion chamber containing the cell culture was perfused with a culture medium. After an exposure period of 24 hours, cell survival was determined by using the MTT assay. Statistical analyses were performed using the Kruskal-Wallis and Mann Whitney U tests.RESULTS: The difference in survival rates between Vertise Flow and Vitrebond groups (positive control) was statistically significant (p<0.05). However, the difference in survival rates between Vertise Flow and President groups (negative control) was not statistically significant (P>0.05).CONCLUSION: Vertise Flow and the negative control group showed similar effects on the survival of the bovine dental pulp-derived cells. Toxic substances released from Vertise Flow may not influence the pulpal cells in case the residual dentin layer is at least 0.5 mm thick.
