Albümin-gluteraldehit doku yapıştırıcısının insan dişeti fibroblast hücreleri üzerindeki etkilerinin incelenmesi

Amaç: Albumin-gluteraldehit doku yapıştırıcısı ilk olarak kardiyovasküler cerrahi uygulamalarında, daha sonra ciddi travmalarda dalak ve kalp yaralanmalarında ve bazı abdominal dokularda, zarar görmüş parankimi güçlendirmek ve/veya hemostaz sağlamak amacıyla geçmişten beri kullanılmaktadır. Diş hekimliğinde ise yönlendirilmiş kemik rejenerasyonu tekniğinde kullanılmaya başlayan albumin-gluteraldehit doku yapıştırıcısının olası komplikasyonlarını tespit etmek amacıyla çalışmamızda, insan dişeti fibroblastı (HGF-1) hücre hattında çene cerrahisi alanında deneysel olarak çalışmaları yapılan albumin-gluteraldehit doku yapıştırıcısı ile klinik uygulamalarda sıklıkla tercih edilen kollajen membranın sitotoksik etkilerini in vitro karşılaştırdık.Gereç ve Yöntem: İki farklı çalışma grubu oluşturularak, 1., 3., 7., 10., 14. ve 21. günlerde albümin-gluteraldehit doku yapıştırıcısından ve kollajen membrandan toplanan süpernatantlar 24, 48 ve 72 saat süreyle insan dişeti fibroblast hücreleri üzerine eklenmiş ve inkübasyon sonrası süpernatantların sitotoksik analizi için MTT yöntemi kullanılmıştır.Bulgular: Tüm ölçümlerde albümin-gluteraldehit doku yapıştırıcısı grubunda hücre canlılık değerinin kollajen membran grubuna göre istatistiksel olarak anlamlı derecede düşük olduğu tespit edilmiştir (p< 0,05). Süpernatantların bekleme süreleri arttıkça hücre canlılık oranlarının azaldığı gözlenmiştir.Sonuç: İnsan dişeti fibroblast hücreleri albumin-gluteraldehit doku yapıştırıcısından elde edilen süpernatantlara karşı sitotoksik davranış göstermiştir.

Investigation of the effects of albumin-gluteraldehyde tissue adhesive on human gingival fibroblast cells

Aim: Albumin-gluteraldehyde tissue adhesive has been firstly used for cardiovascular surgery. After that, it has been applied to spleen, cardiac tissues and abdominal tissues to strengthen parenchyma and to provide hemostasis when damaged in severe trauma. In order to diagnose possible complications of albumin-glutaraldehyde tissue adhesive which has been previously used in guided bone regeneration technique in dentistry, we compared in vitro cytotoxic effects of albumin-glutaraldehyde tissue adhesive that is experimentally used in oral surgery with collagen membrane which is frequently used in clinical practice on human gingival fibroblast (HGF-1) cell line. Material and Methods: Two groups were constituted to investigate cytotoxic effects of supernatants separately collected from albumin-gluteraldehyde tissue adhesive and collagen membrane on 1st, 3rd, 7th, 10th, 14th and 21st days. Supernatants were added to human gingival fibroblast cells for 24, 48 and 72 hours and then, the cytotoxicity was evaluated by MTT analysis.
