Bağ üretim materyalinde kök uru etmeni Agrobacterium tumefaciens'in saptanması

A.tumefaciens biovar 3'e karşı üretilen poliklonal antiserum immunofluoresan testte Orta Anadolu bölgesinde bağ kanseri etmeni A.tumefaciens biovar 3 izolatlarıyla pozitif reaksiyon verdi. Antiserum A.tumefaciens biovar 1 (1/200 seyreltmenin dışında), biovar 2, diğer bitki patojeni ve simbiyotik bakterilerle reaksiyon oluşturmadı. Dormant dönemdeki 1 yaşlı bağ çubuklarının iletim demetlerinde A.tumefaciens"in varlığını saptamak amacıyla vakum ekstraksiyon işlemi uygulandı. Baktirilerin tesbitinde İFAS ve seçici besiyerinden yararlanıldı. Orta Anadolu bölgesinde sağlıklı görünen 150 asmadan 7 sinin, 8 bulaşık asmasının da 6 tanesinin sürgünlerinden A.tumefaciens (biovar 3) izole edildi. IFAS ve seçici besiyerinin etkinliği ve vakum ekstraksiyonunun duyarlılık eşiği deneysel olarak araştırıldı. Aynı zamanda 27 çeşite ait 1 yaşlı aşılı bağ fidanı köklerinde A.tumefacines"m varlığı incelendi. Bu çeşitler¬den 6 tanesinin tümör oluşturan biovar 3, 7 tanesinin de tümör oluşturmıyan biovar 3 veya 1 ile bulaşık olduğu belirlendi. Elde edilen 28 A.tumefaciens izolatından, fizyolojik karakteristiklerine göre 25 tanesi Biovar 3, 3 tanesi de biovar 1 olarak tanılandı.

Detection of crown gal agent (Agrobacterium tumefaciens) in grapevine propagating material

Polyclonal antiserum produced against Agrobacterium tumefaciens (biovar 3) reacted in immunofiuorescent test with bio¬var 3 strains of A.tumefaciens, causal agent of crown gal, from Central Anatolia. The antiserum did not react with A.tumefaciens biovar 1 (except for 1/200 dilution), biovar 2, other plant pathogens and symbionts. Vacuum extraction procedure were applied to recover A.tumefaciens from xylem vessels of one year old dormant grape cuttings. IFAS and selective media were used to detect bac¬teria. A.tumefaciens (biovar 3) was recovered from six cuttings of 8 infected vines and seven of 150 apparently healthy vines in Central Anatolia. The sensitivity thershold of the vacuum extraction method and the effectiveness of the IFAS and selective media were experimentally evaluated. Roots of 1 year old grafted vines of 27 cuitivars were also examined for the presence of A.tumefaciens. Six of these cuitivars were contaminated with tumorigenic biovar 3 and seven of these with nontumorigenic biovar 3 or 1. Of 28 strains of A.tumefaciens isolated, 25 were identified as biovar 3, and 3 as biovar 1 according to their physiological characteristics.
