Pagrus pagrus (L., 1758) yumurtalarının gluteraldehit ile dezenfeksiyonu ve embriyolojik gelişimi
Bu çalışmada, yoğun balık üretimi yapılan kuluçkahanelerde, yumurta ile larval üretime taşınan patojen mikroorganizmalarla mücadele olanakları araştırılarak, dezenfektan olarak kullanılan gluteraldehit (%25’lik, Merck) maddesinin fangri (Pagrus pagrus L., 1758) yumurtalarının embriyolojik gelişimine, açılım miktarına ve bakteri yüküne etkisi incelenmiştir. Yumurtalar 200 ppm glutaraldehit ile 2, 4, 8 ve 16 dak. banyo yoluyla dezenfeksiyon işlemine tabi tutulmuştur. Her denemede 3’er paralel olacak şekilde 1 lt’lik inkübatörlerde günlük olarak yaşama oranları tespit edilmiş, embriyolojik gelişimleri incelenmiş ve yine Petrifilm Flora Total (3M Sante, Fransa) besiyerine 3’er paralelde mikrobiyolojik ekimleri yapılarak bakteri yükleri tespit edilmeye çalışılmıştır. Aynı zamanda 3’er paralelde kontrol grubu (dezenfeksiyonsuz) oluşturularak karşılaştırmalar yapılmıştır. Kontrol gruplarına da deneme gruplarına yapılan tüm işlemler (dezenfeksiyon dışındaki) uygulanarak şartların aynı olması sağlanmıştır. Bu çalışmada en uygun konsantrasyon ve süre (K*S) uygulaması olarak; gluteraldehit ile dezenfeksiyon sonucu 18ºC’de fangri yumurtası için 1600 değeri bulunmuştur. 1600 K*S değerinde tüm gruplarda bakteri gelişimi engellenmiştir. 3200 K*S değerlerinde açılım oranının düştüğü ve süresinin uzadığı bulunmuştur. K*S=1600 değerinden sonra blastomerli safhalarda bozukluklar görülmüştür. Konsantrasyon ve süre değerlerinin sıcaklığa ve türe özgü olarak değişebileceği göz önünde bulundurulursa; tür bazında ve türe özgü sıcaklıklarda denemelerinin yapılarak dezenfeksiyon prosedürlerinin belirlenmesi akuakültür çalışmaları için büyük önem taşımaktadır.

Disinfection of Pagrus pagrus (L., 1758) eggs with glutaraldehyde and embriyological development. In this study, it was investigated to fighting methods with the large number of pathogenic microorganisms which transferred the larval production from brood stock, and effect of the glutaraldehyde (Merck, 25%) that was used as a disinfectant to red porgy (Pagrus pagrus L., 1758) embryological development, the hatching rate and the bacterial flora were searched. The eggs were disinfected with 200 ppm glutaraldehyde; 2, 4, 8, 16 min. at four different contact times. The daily hatching rate and embryological development were determined with 3 triplicates for every treatment, and bacteria colonies were counted by Petrifilm Flora Total (Laboratoire 3M Sante, France) agar planting, as well. At the same time the control groups were established at triplicate and were made the comparisons. The control groups (without treatment) were included in all trials. In this study, it is found that the optimum C*T treatment is 1600 for red porgy eggs at 18ºC. In all 1600 C*T treatments the bacterial growth were inhibited. In all 3200 K*S treatments the hatching rates were decreased and hatching time was delayed. After C*T=1600 values it was observed some morphological problems after 8 blastomer stages. Considering the C*T values change with the species and the temperature, it is very important the determination of the disinfection procedures for different species on aquaculture.
