Karadeniz'de yaşayan Pisi Balığı'nın Platichthys flesus luscus (Pallas, 1814) (Pisces, Pleuronectiformes) ilk karyolojik tanımlaması

Bu çalışmada, Karadeniz'de yaşayan yassı balık türlerinden (Pleuronectiformes) birisi olan Platichthys flesus luscus (Pallas, 1814) (Pisi Balığı)'un kromozom yapıları, sayıları ve sitogenetik özellikleri çeşitli bantlama yöntemleri kullanılarak incelenmiştir. İncelenen balık örnekleri Batı ve Orta Karadeniz sahili boyunca (Zonguldak ve Ordu illeri arası) 5 bölgeden avcılıkla elde edilmiştir. Büyük balıklara (250-1500 g) % 0,05 kolşisin solüsyonu (25-50 µg/g vücut ağırlığı) dorsalden kas arasına, ventralden karın boşluğuna enjekte edilmiştir. Küçük balıklar (50-250 g) ise % 0,005 kolşisin solüsyonunda 6 saat süre ile canlı bekletilmişlerdir. Diseksiyonla çıkarılan solungaç ve yüzgeç epitel dokuları % 0,4 KCl solüsyonunda 30 dk. bekletilip, Carnoy fiksatifi ile muamele edilmiştir. Kuruyan preparatlar en az 15 dakikalık süreyle giemsa (pH 6,8) solüsyonunda boyanmıştır

The first caryological description of European Flounder, Platichthys flesus luscus (Pallas, 1814) (Pisces, Pleuronectiformes) living in the Black Sea

In this study, the chromosome structures, numbers and other cytogenetic features of flounder, Platichthys flesus luscus (Pallas, 1814), which is one of the flatfish species in the Black Sea, were examined with using different banding methods. Fish samples were obtained from fishermen in 5 localities between Zonguldak and Ordu of the West and Middle Black Sea coasts. A total of 0.05 % of colchicine solution (25-50 µg / g body) weight was injected from intramuscular to dorsal and from intraperitonal to ventral for big fish (body weight 250 to 1500 g). However, small fish (body weight 50 to 250 g) were maintained in 0.005 % of colchicine solution for 6 hours. Gill and fin epithelia tissues derived by dissecting were incubated in 0.4 % of KCl solution for 30 mins and treated with Carnoy's fixative. Dried slides were stained with giemsa (pH 6.8) solution for 15 mins.
