SIĞIRLARDA EMBRİYO TRANSFERİNDE CIDR İLE SENKRONİZE EDİLEN DONÖRLERE ÖSTRUS ÖNCESİ GERÇEKLEŞTİRİLEN ÇİFT PGF2α UYGULAMALARININ ELDE EDİLEN EMBRİYOLARIN KALİTESİ VE SAYISI ÜZERİNE ETKİLERİ
Bu çalışmada, sığırlarda embriyo transferinde östrus öncesi uygulanan çift PGF2α uygulamalarının elde edilen embriyoların kalitesi ve sayısı üzerine etkileri araştırıldı. Araştırmada, 5-7 yaşlı Holştayn ırkı, herhangi bir sağlık ve reprodüktif problemi bulunmayan, siklusları düzenli seyreden 20 baş Holştayn ırkı inek iki gruba ayrıldı (n=10). Donörlere östrus siklusunun herhangi bir gününde CIDR uygulandı. CIDR uygulandıktan sonra süperovulasyon amacıyla yedinci günden başlayarak dört gün süreyle 12 saat aralıklarla azalan dozlarda (80:80 mg, 60:60mg, 40:30 mg, 30:20 mg) toplam 400 mg FSH kas içi yolla uygulandı. Daha sonra corpus luteumu (CL) lize etmek amacıyla Grup I’de 5. FSH enjeksiyonuyla birlikte tek doz 500 μg Cloprostenol ve Grup II’de 5. ve 6. FSH enjeksiyonuyla birlikte çift doz toplam 1000 μg kas içi enjekte edildi. Her iki grupta da 6. FSH enjeksiyonuyla birlikte CIDR uzaklaştırıldı. Cloprostenol enjeksiyonundan 24 saat sonra günde üç kez donörlerin östrusları takip edildi ve östrus başlangıcından itibaren 12 saat ara ile üç kez tohumlandı. Embriyolar tohumlamayı takiben yedinci günün sonunda uterus yıkaması ile toplandı. Sonuç olarak, iki protokol arasında embriyo sayısı ve kalitesi açısından fark bulunamamıştır. Ancak donörlere östrustan önce çift prostaglandin uygulamasının embriyo sayısı ve kalitesi üzerine sayısal olarak olumlu etkisinin olduğu kanaatine varılmıştır
The Effect of Double Pgf2α Administrations Applıed to Donor Before Oestrus With Synchronized Cidr on the Number and Qualıty of Obtained Embryos DuringEmbryo Transfer in Cattle
In this study, the effect of embryo transfer applications from pre-applied double-PGF2α before the oestrus period on the number and quality of the embryos in cattle were investigated. In the study, 20 Holstein cows aged 5 to 7 years and do not have any health and reproductive problems and had regular cycles were divided into two groups (n = 10). CIDR was applied to donors in any one day of estrous cycle. İn order to superovulation decreasing FSH doses were intramuscularly administered after CIDR application by starting from 7th day with 12-hour intervals for four days (mg/ 80:80 mg, 60:60 mg, 40:30 mg, 30:20 mg) which was totally 400 mg. And later, in order to lyses corpus luteum with 5th FSH injections with a single dose 500 μg Cloprostenol and with 5th and 6th FSH injections with a double dose (500 μg+500 μg) Cloprostenol intramuscularly injected to Group I and Group II, respectively. In both Groups, CIDR was removed at the same time with 6th FSH injections. Donors’ oestrus were followed up after 24 hours of cloprostenol injection three times a day and from the beginning of estrus with a 12-hours intervals three times inseminated. Embryos were collected at the end of the 7th day following the insemination, by uterus washing (flush). In conclusion, no statistically significant differences between the two protocols was found in terms of embryo quality and number, but it was concluded that double-prostaglandin applications to the donors’ before the oestrus, had positive impact on the quality and number of embryos.
