Sesamum indicum L. ve Apium graveolens L. ekstreleri karboplatin hepatotoksisitesine karşı koruyucu mu?

Amaç: Bu çalışmada karboplatinin sıçan karaciğerinde yaptığı toksisite üzerine Sesamum indicum L. ve Apium graveolens L. ekstrelerinin koruyucu etkileri araştırıldı. Yöntem: Sprague-Dawley ırkı sıçanlardan oluşturulan gruplara serum fizyolojik (SF), karboplatin, karboplatin+Sesamum indicum L. (susam) sabit yağı (SSY), karboplatin+Apium graveolens L. (kereviz) sabit yağı (KSY), karboplatin+C vitamini ve karboplatin+E vitamini uygulandı. Bulgular: Çalışma gruplarına ait karaciğerlerde histopatolojik bulguya rastlanmadı. Serum ALT, ALP ve indirekt bilirubin seviyeleri karboplatin grubunda SF kontrol grubuna göre anlamlı derecede yüksekti ve karboplatinin biyokimyasal enzimler düzeyinde karaciğer hasarı yaptığı şeklinde yorumlandı. SSY ve KSY’nin ALT, ALP, AST ve indirekt bilirubin düzeyleri karboplatin grubundan anlamlı derecede düşüktü. Yalnızca SSY grubunun ALP değeri SF kontrol grubundan yüksek olarak tespit edildi, bunun dışında SSY ve KSY ile SF kontrol grubu arasında anlamlı bir farklılık saptanmadı. C vitamini ve E vitamini gruplarına ait biyokimyasal değerler de SSY ve KSY’na benzer şekilde bulundu. SF, karboplatin ve SSY gruplarının günlük vücut ağırlığı değişimi birbirine benzer seyretti. KSY, C vitamini ve E vitamini gruplarının günlük vücut ağırlığı değişimi ise gittikçe azalan bir seyir gösterdi. Sonuç: SSY ve KSY’nin karboplatine bağlı karaciğer toksisitesini kısmen önleyebileceği sonucuna varıldı.

Are fixed oils of Sesamum indicum L. and Apium graveolens L. protective against hepatotoxicity?

Objective: Effects of Sesamum indicum L. and Apium graveolens L. extracts against carboplatin-induced hepatotoxicity were aimed to investigate in rats in this study. Method: Sprague-Dawley rats were divided into six groups and given isotonic saline solution (ISS), carboplatin, carboplatin+fixed oil of Sesamum indicum L. (FOS), carboplatin+fixed oil of Apium graveolens L. (FOA), carboplatin+vitamin C and carboplatin+vitamin E, respectively. Results: In histological examination of liver sections none of the groups exhibited pathological changes. The liver injury was evidenced by rise of serum ALT, ALP and indirect bilirubin levels in carboplatin group compared to the controls. The FOS and the FOA groups displayed significantly lower ALT, ALP and indirect bilirubin levels compared to the carboplatin group. The only difference from the ISS group was elevation of ALP level in FOS. Vitamin C and E displayed similar biochemical changes as FOS and FOA. Body weights were similar in ISS, carboplatin and FOS groups throughout the study but progressively decreased in FOA, vitamin C and vitamin E groups. Conclusion: FOS and FOA appears to be partially effective in alleviating carboplatin-induced hepatotoxicity.
