Bromodeoksiuridin uygulamasını takiben erişkin sıçan beyninde lateral ve üçüncü ventriküllerin ventriküler ve subventriküler zonunda mitojenik aktivitenin incelenmesi

Amaç: Çalışmamızın amacı erişkin sıçanların subventriküler zonunda (SVZ) veya ventriküler zonunda (VZ) bromodeoksiuridin (BrdU) işaretli yeni oluşan hücreleri saptamaktı. Yöntem: 13-14 haftalık erişkin Spraque Dawley cinsi (n=7) erkek sıçanlara BrdU enjekte edildi ve enjeksiyondan 24 saat sonra perfüzyon yapıldı. Mitojenik aktiviteyi saptamak için BrdU antikor immünohistokimyası kullanıldı ve BrdU işaretli hücreler mikroskopta sayıldı. Bulgular: Lateral ve üçüncü ventriküllerin VZ’undaki ependimal hücrelerde BrdU işaretli nukleus gözlenmedi. Lateral ve üçüncü ventriküllerin SVZ’da ise BrdU işaretli nukleuslar gözlendi. In the brain of the adult rat following applicatıon bromodeoxiuridine examine mitogenic activity in the ventricular and the subventricular zone of the lateral and the third ventricle

Objective: The aim of our study was to determine bromodeoxyuridine (BrdU) labeled newborn cells in the subventricular zone (SVZ) or ventricular zone (VZ) of adult rats. Methods: 13-14 weeks old Spraque Dawley (n=7) male rats were injected with BrdU and were perfused 24 hours after the injection. We used BrdU antibody immunohistochemistry to determine that mitogenic activity and BrdU labeled cells counted under microscope. Results: In the ependymal cells at VZ of lateral and 3rd ventricles were not seen BrdU labeled nucleus. In contrast, in the SVZ of lateral and 3rd ventricles were seen BrdU labeled nucleus. In the SVZ of lateral ventricles was seen BrdU labeled nucleus more significantly different than in the SVZ of 3rd ventricles with the statistical analysis (P<0.001). Conclusion: In the VZ of lateral and 3rd ventricle was not detected mitotic activity. In contrast, in the SVZ of lateral and 3rd ventricle was detected mitotic activity. In the SVZ of lateral ventricles was detected more mitoticaly active than in the SVZ of 3rd ventricles.
