Molecular analysis of Turkish Alyssum L. (Brassicaceae) species by RAPD-PCR and SDS-page methods

Türkiye'de yetişen ve nikel metalini yaprakta biriktirme özelliği nedeni ile ekolojik değeri olan bazı Alyssum L. (Brassicaceae) türleri arasındaki genetik uzaklıklar RAPD-PCR ve SDS-PAGE yöntemleri ile belirlenmiştir. RAPD-PCR çalışmasında toplam 42 primer denenmiştir. Bunlardan beş primer polimorfik bantlar vermiş ve OPI18 primeri genetik uzaklığın tanımlanması için seçilmiştir. Genetik uzaklıklar hem RAPD-PCR hem de SDS-PAGE yöntemlerinde çalışılan türler için hesaplanmış ve SPSS bilgisayar programı ile dendrogramlar çıkartılmıştır. Alyssum cinsine ait türlerden SDS-PAGE yöntemi ile incelenenlerde türler arası polimorfizm, RAPD-PCR yöntemi ile incelenenlerde ise tür içi polimorfizm varlığı tespit edilmiştir.

Türkiye' nin bazı Alyssum L. (Brassicaceae) türlerinin RAPD-PCR ve SDS- page yöntemleri ile moleküler analizleri

RAPD-PCR and SDS-PAGE were used to investigate the pattern of genetic variation among some species of Alyssum L.(Brassicaceae); a nickel hyperaccumulator plant growing in Turkey. Forty two primers were used for the RAPD-PCR analyses. Polymorphic bands were obtained with five primers and OPI18 was selected for identifying the genetic distances. Genetic distances were calculated for all the species studied at both RAPD-PCR and SDS-PAGE methods, dendrograms were obtained with the SPSS computer program. Polymorphisms between Alyssum species and within species were determined by using SDS-PAGE and RAPD-PCR, respectively.
